1. Cell Structure and Organelles |
One common mistake is confusing the cell wall with the cell membrane, especially in plant and bacterial cells. The cell wall is a rigid outer layer made of cellulose (plants) or peptidoglycan (bacteria) that provides structural support, while the cell membrane is a selectively permeable barrier controlling substance movement. Exam questions often ask you to label diagrams of a typical plant cell, animal cell and bacterial cell, so memorise the key differences: plant cells have a permanent vacuole, chloroplasts and a cell wall; animal cells have none of these; bacterial cells have circular DNA, plasmids, no nucleus and usually a cell wall.
Another pitfall is misidentifying organelles on electron micrographs. Students often label mitochondria as chloroplasts because of the folded inner membranes, but mitochondria have cristae, while chloroplasts contain stacks of thylakoids called grana. Remember that both plant and animal cells contain mitochondria; chloroplasts are only in photosynthetic plant cells and some protists.
When comparing eukaryotes and prokaryotes, avoid saying prokaryotes have no DNA — they lack a nucleus, but genetic material is present as a circular chromosome and plasmids. Also, scale is vital: bacterial cells are generally much smaller than eukaryotic cells, and examiners may test this using orders of magnitude calculations.
2. Diffusion, Osmosis and Active Transport |
These three transport mechanisms are among the most heavily examined topics. The number one error is treating osmosis simply as 'the movement of water'. A full-mark definition needs: "the net movement of water molecules from a region of higher water potential to a region of lower water potential across a partially permeable membrane." Many students omit "net", "water potential", or "partially permeable membrane", all of which are essential for marks.
Diffusion is the net movement of particles from a region of high concentration to a region of low concentration, down a concentration gradient, and is passive. Active transport, by contrast, requires energy from respiration to move substances against their concentration gradient, using carrier proteins. Confusing which processes need energy is a classic mistake: both diffusion and osmosis are passive, while active transport and processes like endocytosis are active.
When investigating osmosis using potato cylinders or visking tubing, common errors include not blotting the cylinders properly before measuring final mass, or using concentrations of sugar/salt outside a suitable range, leading to non-linear results. Always list control variables: temperature, volume of solution, surface area of tissue, and time. Be precise about how you measure mass change and calculate percentage change to allow fair comparison.
3. Enzymes and Factors Affecting Enzyme Activity |
Students often describe enzymes as being 'killed' by high temperature or extreme pH. The correct scientific term is 'denatured'. Denaturation is a permanent change to the active site's shape, preventing the substrate from binding. The lock-and-key model helps explain specificity: only a substrate with a complementary shape fits the active site. Modern specifications also reference the induced-fit model, where the active site changes shape slightly as the substrate binds, which you should mention for top marks.
In graph analysis, a common error is misreading the plateau of an enzyme‑controlled reaction. The plateau occurs not because the enzyme is denatured, but because the substrate concentration becomes a limiting factor — all active sites are occupied. Another trick: when temperature is increased, the initial rate rises due to more frequent collisions, but beyond the optimum, the rate drops sharply as enzymes denature. Practise describing the shape of the curve in terms of kinetic energy and denaturation.
Required practical: effect of pH on amylase activity. Pupils often forget to allow the starch-iodine colour change to progress to a set end-point, or they misuse water baths and fail to control temperature. Also, when testing for reducing sugars using Benedict's solution, remember it is a qualitative test — a colour change from blue to green, yellow, orange, or brick-red indicates the presence of reducing sugar; a lack of change indicates absence. Never say it gives a definitive concentration unless you are using a colorimeter.
4. Digestive System and Enzyme Actions |
A high-frequency topic is the site of production and action of digestive enzymes. Amylase is produced in the salivary glands and pancreas, and works in the mouth and small intestine to break down starch into maltose. Proteases are produced in the stomach (pepsin), pancreas (trypsin) and small intestine; they convert proteins into amino acids. Lipases are produced in the pancreas and act in the small intestine, breaking lipids into fatty acids and glycerol. Common error: believing the stomach produces lipase — it does not; the stomach's acidic environment is for protein digestion by pepsin.
Bile is often misunderstood. It is produced by the liver, stored in the gall bladder, and released into the small intestine. Bile does not contain enzymes; it emulsifies fats, breaking large lipid droplets into smaller ones to increase surface area for lipase action. Also, bile is alkaline, so it neutralises stomach acid, providing an optimum pH for pancreatic and intestinal enzymes. Stating that bile 'digests fats' will lose marks.
The villi of the small intestine provide an excellent example of adaptation. Exam answers should refer to large surface area (folds and microvilli), thin epithelium (one cell thick) for short diffusion distance, good blood supply to maintain a concentration gradient, and lacteals for lipid absorption. A typical error is forgetting that absorbed glucose and amino acids enter the blood capillaries, while fatty acids and glycerol enter the lacteals.
5. The Heart and Circulatory System |
Heart structure questions regularly trap students. The right side of the heart pumps blood to the lungs (pulmonary circulation) and the left side pumps to the body (systemic circulation). The left ventricle has a much thicker muscular wall because it must generate higher pressure to push blood all around the body. Mixing up left and right on a diagram is extremely common — remember that left and right are labelled as if the heart were inside your own chest, facing you.
lood: vena cava → right atrium → right ventricle → pulmonary artery → lungs → pulmonary veins → left atrium → left ventricle → aorta. Students often reverse the pulmonary artery and pulmonary vein — ngs). The pulmonary artery is the only artery carrying deoxygenated blood; the pulmonary vein is the only vein carrying oxygenated blood.
In terms of blood vessels, arteries have thick elastic and muscular walls to withstand high pressure; capillaries are one-cell thick for exchange; veins have valves and a larger lumen. For exam success, link structure to function in every description. A common misconception is that all arteries carry oxygenated blood — as explained above, the pulmonary artery carries deoxygenated blood to the lungs.
6. Respiration: Aerobic and Anaerobic |
rom glucose, which is used to synthesise ATP. A common exam blunder is calling respiration 'breathing' or saying its purpose is 'to produce energy' en → carbon dioxide + water. Be able to write the balanced chemical symbol equation too: C₆H₁₂O₆ + 6O₂ → 6CO₂ + 6H₂O.
Anaerobic respiration in animal cells produces lactic acid and much less ATP than aerobic respiration. In yeast and some plants, anaerobic respiration produces ethanol and carbon dioxide — this is the basis for fermentation in bread-making and brewing. Pupils frequently confuse the products: animals do not produce ethanol. Memorising which organisms produce which products is a must. Oxygen debt is a related concept: after vigorous exercise, extra oxygen is needed to break down accumulated lactic acid in the liver.
In experiments to measure respiration rate, such as using a respirometer with germinating seeds, a frequent error is forgetting the role of soda lime (to absorb carbon dioxide) so that changes in gas volume are due solely to oxygen consumption. Without it, pressure changes could be misinterpreted. Also, describe the need for a thermostatically controlled water bath to maintain constant temperature, because temperature affects gas volume.
7. Photosynthesis and Plant Nutrition |
e presence of light and chlorophyll. A recurring mistake is thinking that oxygen comes from carbon dioxide — plit during the light-dependent reactions (photolysis). Many marks are lost by leaving out 'chlorophyll' or 'light' as essential conditions.
Limiting factors: light intensity, carbon dioxide concentration and temperature. The graph for light intensity is often misinterpreted. The plateau occurs because something else (e.g., CO₂ or temperature) becomes the limiting factor. Students should be able to design an experiment to measure the effect of light intensity on the rate of photosynthesis using Canadian pondweed, counting oxygen bubbles. Key pitfalls: not placing the lamp at a fixed distance, not allowing the plant to equilibrate, or ignoring the heat produced by the lamp which affects temperature — a heat shield or LED lamp is needed.
Leaf structure is another staple. Label the waxy cuticle (prevents water loss), upper epidermis, palisade mesophyll (densely packed with chloroplasts for maximum light absorption), spongy mesophyll (air spaces for gas exchange), lower epidermis with guard cells and stomata. A mistake is to state that chloroplasts are found in all leaf cells — they are mainly in palisade cells and guard cells.
Transpiration is the loss of water vapour from leaves via stomata. Factors affecting rate include temperature, humidity, wind speed and light intensity. Using a potometer to measure water uptake: common mistake is saying it measures transpiration rate directly — it actually measures water uptake, which is almost equal to transpiration rate if the plant is healthy. Always cut the stem underwater to prevent air locks and mention that the air bubble must be allowed to move past the start mark.
8. Endocrine System and Hormonal Control |
The menstrual cycle and hormonal control is a guaranteed high-mark question. Follicle-stimulating hormone (FSH) from the pituitary stimulates follicle development and oestrogen production. Oestrogen from the ovary repairs and thickens the uterine lining and stimulates a surge in luteinising hormone (LH). LH triggers ovulation and stimulates the formation of the corpus luteum, which secretes progesterone to maintain the uterine lining. If no pregnancy occurs, progesterone levels fall, causing menstruation. Common mistake: mixing up FSH and LH functions or saying progesterone is produced by the pituitary — it is produced by the corpus luteum in the ovary.
Blood glucose regulation is another favourite. Insulin is secreted by the pancreas when blood glucose is too high, causing cells to absorb more glucose and the liver to convert glucose to glycogen for storage. Glucagon is secreted when blood glucose is too low, causing the liver to break down glycogen back into glucose. The pancreas also detects the blood glucose level directly. The common slip-up is calling this a negative feedback mechanism but failing to explain it: explain how a change from the set point triggers a response that counteracts the change, returning the system to normal.
Type 1 and Type 2 diabetes are often contrasted. Type 1 is an autoimmune condition where the pancreas produces little or no insulin, usually from a young age, treated with insulin injections. Type 2 is linked to obesity and lifestyle, where body cells become resistant to insulin, often managed by diet, exercise, and sometimes medication. Do not say Type 1 is 'caused by eating too much sugar'.
9. Human Reproduction and Contraception |
The structure and function of male and female reproductive systems are frequently examined with labelling tasks. In females, the fallopian tubes (oviducts) transport the egg to the uterus; fertilisation normally occurs in the fallopian tube. In males, sperm are produced in the seminiferous tubules of the testes and stored in the epididymis. The prostate gland and seminal vesicles add fluids to nourish sperm and aid swimming. Avoid confusing the scrotum's function (keeping testes cooler) with the testes' primary role (sperm and testosterone production).
Hormonal methods of contraception include oral contraceptives containing oestrogen and progesterone, implants, injections, and the IUD (which can also be copper-based and non-hormonal). Barrier methods like condoms and diaphragms prevent sperm from reaching the egg. The common exam error is neglecting to discuss advantages and disadvantages. For full marks, you need to evaluate a method in terms of effectiveness, side effects, reversibility and protection against sexually transmitted infections.
10. DNA, Genetics and Inheritance | DNA、
The structure of DNA is a double helix, with sugar-phosphate backbones and complementary base pairing: adenine with thymine (A-T), cytosine with guanine (C-G). Nucleotides consist of a sugar, a phosphate group and a base. A common mistake is writing A-C and T-G pairings, or saying that DNA is made of amino acids — it is a nucleic acid. The genome is the entire genetic material of an organism.
Genetic crosses start with key definitions: dominant, recessive, homozygous, heterozygous, genotype and phenotype. When using Punnett squares, always state the parental genotypes, possible gametes, offspring genotypes and phenotypes, and express the ratio clearly. The major pitfall is misinterpreting ratios when inheritance is sex-linked, such as haemophilia or colour blindness carried on the X chromosome. Also, do not write gametes with two alleles — each gamete contains only one allele for each gene.
Monohybrid crosses involving pedigree analysis often appear. You need to deduce genotypes from family trees and calculate probabilities. Remember that an affected child can be born to unaffected parents if both are heterozygous carriers for a recessive condition. Always explain reasoning step by step. A common slip is using the term 'carrier' for dominant disorders — carriers exist only for recessive conditions.
11. Evolution by Natural Selection |
The mechanism of natural selection can be summarised: within a population there is genetic variation; individuals with advantageous alleles are better suited to their environment and more likely to survive, reproduce and pass on those alleles to their offspring; over many generations, the advantageous allele becomes more common in the population. The common mistake is saying that organisms 'adapt' during their lifetime or that they 'choose' to evolve. Natural selection acts on existing variation — it does not create new traits on demand.
Antibiotic resistance in bacteria is a classic example. When exposed to antibiotics, most bacteria die, but some with a resistance gene survive. These survivors reproduce, and the resistance gene spreads. Pupils often incorrectly state that antibiotics 'cause' the mutation. In reality, the mutation occurs randomly and the antibiotic acts as a selective agent. Emphasise the importance of finishing a prescribed course of antibiotics to reduce the chance of resistant strains evolving.
Speciation is the formation of a new species through isolation. Two populations of the same species become geographically separated, preventing gene flow. Different environments exert different selection pressures, so different alleles become advantageous. Over time, the populations diverge so much they can no longer interbreed to produce fertile offspring. A typical error is claiming that speciation occurs in one or two generations — it requires many generations.
12. Ecosystems, Cycles and Biodiversity |
Food chains and energy transfer are straightforward but often sloppy in exams. Energy is transferred along trophic levels, but only about 10% is passed on — the rest is lost as heat through respiration, or in uneaten parts and waste. A common mistake is forgetting to mention that energy is lost to the environment as heat, or that decomposers use organic matter as a food source. Pyramids of biomass are generally regular, but pyramids of numbers can be inverted, e.g., one large tree supports many insects.
The carbon cycle requires you to describe how carbon dioxide is removed from the atmosphere by photosynthesis, how it is returned by respiration, combustion and decomposition. The role of microorganisms in decay is critical — they release enzymes to decompose dead matter and return mineral ions to the soil. Many students lose marks by mixing up nitrogen and carbon cycles. Remember: nitrifying bacteria convert ammonia to nitrates, nitrogen-fixing bacteria convert atmospheric nitrogen into compounds plants can use, and denitrifying bacteria return nitrogen to the atmosphere. These names are often confused.
Biodiversity and sustainable practice questions often ask for consequences of deforestation or fishing. Deforestation reduces biodiversity, destroys habitats, affects the water cycle and releases carbon dioxide if trees are burned. Overfishing reduces fish stocks below sustainable levels and disrupts food webs. Conservation methods include setting quotas, using larger net mesh sizes to allow young fish to escape, and replanting trees. When evaluating, always give both positives and negatives to show balanced thinking.
