📚 AQA Chemistry: Nucleophilic Substitution SN1 and SN2 and Elimination Mechanisms | AQA

ctical technique inside a single area of organic chemistry. A halogenoalkane contains a polar carbon-halogen (C-X) bond, so the carbon atom carries a partial positive charge, δ⁺, and is attacked by nucleophiles. Two pathways compete: nucleophilic substitution replaces the halogen with another group, while elimination removes the halogen together with a hydrogen from the neighbouring carbon to form a C=C double bond. Which pathway dominates depends on the class of halogenoalkane (primary, secondary or tertiary), the reagent, the solvent and the temperature.

1. Why This Topic Carries So Many Marks |

Examiners reward three separate skills here. First, you must be able to draw a mechanism accurately, using curly arrows that start from a lone pair or a bond and end at the atom that accepts the electrons. Second, you must be able to select the correct conditions for substitution or elimination and justify the choice. Third, you must be able to explain trends in reactivity using bond enthalpy and structure. The topic also feeds directly into later work on alcohols, alkenes, amines, nitriles and polymers, so a weak understanding here causes problems across the whole organic paper.

AQA expects you to know the mechanism of nucleophilic substitution for a primary halogenoalkane and for a tertiary halogenoalkane, and the mechanism of elimination for a halogenoalkane with ethanolic potassium hydroxide. Everything in this article sits inside specification sections 3.3.1.4 and 3.3.1.5, and each idea below appears regularly in Paper 2 and Paper 3.

2. The Carbon-Halogen Bond: Polarity versus Bond Enthalpy |

Everything starts with the C-X bond. Halogens are more electronegative than carbon, so the shared pair is pulled towards the halogen. The halogen therefore becomes δ⁻ and the carbon becomes δ⁺, creating the electrophilic centre that a nucleophile attacks. Two trends matter, and they pull in opposite directions. Polarity decreases from C-F to C-I as electronegativity falls, but bond enthalpy also falls sharply down the group, so the C-I bond is by far the weakest.

Halogen | 卤素Approx. bond enthalpy of C-X / kJ mol⁻¹ | C–X 键焓(约)Polarity of C-X | C–X 极性Relative rate of hydrolysis | 水解相对速率

The rate of hydrolysis is governed mainly by bond enthalpy, not by polarity, so the order is C-I > C-Br > C-Cl > C-F. This is one of the most frequently tested traps in the whole specification: the most polar bond reacts slowest, because breaking it costs the most energy.

3. Nucleophilic Substitution: The Overall Picture |

In nucleophilic substitution, a nucleophile donates a lone pair to the δ⁺ carbon while the halogen leaves as a halide ion. The general equation for hydrolysis with aqueous alkali is shown below, and a typical example follows it.

R-X + OH⁻ → R-OH + X⁻

CH₃CH₂Br + OH⁻ → CH₃CH₂OH + Br⁻

The standard conditions are warm aqueous sodium or potassium hydroxide under reflux. The word "aqueous" is essential: water is present as the solvent and as the source of the hydroxide ions, and it also stabilises the leaving halide ion. The product is an alcohol. Note that the hydroxide ion acts as both a nucleophile and a base, so the same reagent can in principle give two different products depending on conditions.

The organic product is an alcohol in which the OH group sits on the carbon that previously carried the halogen. If you need to lengthen a carbon chain rather than introduce an OH group, you switch the nucleophile to cyanide, which is covered later in this article.

4. The SN2 Mechanism: Concerted Bimolecular Attack | SN2

SN2 stands for substitution, nucleophilic, second order. The mechanism is a single concerted step: the nucleophile attacks the δ⁺ carbon from the side opposite the halogen at the same moment that the C-X bond breaks. There is no intermediate, only a transition state in which the carbon is partially bonded to five groups and carries a partial negative charge, while the leaving halogen also carries a partial negative charge.

CH₃Br + OH⁻ → [HOδ⁻···CH₃···Brδ⁻]‡ → CH₃OH + Br⁻

Because both species appear in the rate-determining step, the rate equation contains both concentrations, and the reaction is first order with respect to each of them. Any change in the concentration of either the halogenoalkane or the hydroxide ion changes the rate.

rate = k[R-X][OH⁻]

SN2 is favoured by primary halogenoalkanes, because the carbon bearing the halogen has only one alkyl group, so there is little steric hindrance and the nucleophile can approach from the back with ease. Tertiary halogenoalkanes are effectively excluded from this pathway: three bulky alkyl groups block the approach completely.

5. The SN1 Mechanism: Unimolecular and Carbocation-Based | SN1 机理:单分子与碳正离子

SN1 stands for substitution, nucleophilic, first order. The reaction happens in two steps. In the slow, rate-determining step the C-X bond breaks heterolytically, with both bonding electrons going to the halogen, and a carbocation intermediate forms. In the fast second step the nucleophile attacks the positively charged carbon.

(CH₃)₃C-Br → (CH₃)₃C⁺ + Br⁻   (slow | 慢)

(CH₃)₃C⁺ + OH⁻ → (CH₃)₃C-OH   (fast | 快)

Only the halogenoalkane is involved in the slow step, so the rate equation contains just one concentration and the overall order is one. Adding more hydroxide makes the second step faster but does not change the measured rate.

rate = k[R-X]

SN1 is favoured by tertiary halogenoalkanes because the resulting carbocation is strongly stabilised. Three alkyl groups push electron density towards the positive centre by the inductive effect, and additional stabilisation comes from hyperconjugation. Primary carbocations are far too unstable to form, which is why primary halogenoalkanes never react by SN1.

6. Evidence and Rate Equations: Telling SN1 from SN2 | 证据与速率方程:区分 SN1 与 SN2

Kinetic evidence is the cleanest way to decide which mechanism is operating. If doubling the concentration of the halide ion source has no effect on the rate, the hydroxide is not in the rate-determining step, so the mechanism must be SN1. If doubling the nucleophile concentration doubles the rate, both species are in the slow step, so the mechanism is SN2. Structural evidence supports this: primary halogenoalkanes react by SN2 and tertiary by SN1, with secondary halogenoalkanes sitting on the borderline and depending on conditions.
