📚 CIE A-Level Organic Chemistry: Mechanisms and Study Guide | CIE A-Level

Organic chemistry is a cornerstone of the CIE A-Level Chemistry syllabus, demanding both conceptual understanding and the ability to apply reaction mechanisms. Mastering mechanisms is not just about memorising equations; it is about recognising electron flow, understanding the role of reagents, and predicting products. This guide provides a structured overview of the key reaction mechanisms, together with practical study strategies to help you excel in Paper 2 (AS Structured Questions), Paper 4 (A2 Structured Questions) and Paper 5 (Planning, Analysis and Evaluation).

1. Functional Groups and Reactivity |

nal groups are the reactive centres of organic molecules. Common groups include alkanes (C–C, C–H), alkenes (C=C), halogenoalkanes (C–X), alcohols (–OH), aldehydes (–CHO), ketones (C=O with two carbon substituents), carboxylic acids (–COOH), esters (–COOR), acyl chlorides (–COCl), amines (–NH₂) and nitriles (– ctivity dictated by bond polarity, and the presence of lone pairs, or delocalised π electrons.

A useful way to predict reactivity is to ask two questions: is there an electron-rich site (lone pair, π bond, or carbanion), and is there an electron-poor site (polarised bond with a δ⁺ carbon)? Reactions happen when these two sites meet. The polarity of a bond depends on electronegativity difference: C–O, C–Cl and C–N bonds are polar, whereas C–C and C–H bonds are essentially non-polar.

Functional group Reactive feature Typical mechanism
C=C (alkene) π electrons, electron-rich Electrophilic addition
C–X (halogenoalkane) Polar bond, δ⁺ carbon Nucleophilic substitution / elimination
C=O (carbonyl) Polar bond, δ⁺ carbon Nucleophilic addition
Arene ring Delocalised π electrons Electrophilic substitution

2. Reagents, Bond Fission and Curly Arrows |

Reagents are classified by what they attack. A nucleophile is an electron-pair donor (OH⁻, CN⁻, NH₃, H₂O, Br⁻, the π bond of an alkene) and attacks electron-poor centres. An electrophile is an electron-pair acceptor (H⁺, NO₂⁺, Br⁺, AlCl₃, the δ⁺ carbon of a carbonyl) and attacks electron-rich centres. A radical contains an unpaired electron and reacts to pair it up.

Bonds break in two ways. Homolytic fission splits a bond evenly, giving two radicals, each with one electron of the pair; it is favoured by UV light or high temperature and by non-polar bonds. Heterolytic fission splits a bond unevenly, giving a cation and an anion; it is favoured by polar solvents and by polar bonds. Organic mechanisms at A-Level are almost always heterolytic, except for free radical substitution.

Curly arrows show the movement of an electron pair. The arrow always starts from the source of electrons (a lone pair, a σ bond or a π bond) and points to where the pair goes — usually an atom, forming a new bond. A full arrow (two barbed heads) means two electrons; a half-headed "fish-hook" arrow means one electron and is only used in radical mechanisms. Marks in CIE exams are awarded for arrows starting from the correct place and ending at the correct atom, so precision matters more than artistic skill.

3. Nucleophilic Substitution: The SN2 Mechanism | 亲核取代:SN2 机理

SN2 stands for substitution, nucleophilic, bimolecular. The nucleophile attacks the carbon bearing the halogen from the side opposite the leaving group, in a single concerted step. The C–X bond breaks as the C–Nu bond forms, and the transition state has five groups around the carbon. The rate depends on both reactants:

rate = k[CH₃CH₂Br][OH⁻]

Because the nucleophile must approach from the back, the reaction is sensitive to steric hindrance: primary halogenoalkanes react fastest, tertiary ones essentially not at all by SN2. Backside attack also inverts the configuration at the carbon, a result known as Walden inversion. SN2 is favoured by a strong nucleophile (OH⁻, CN⁻, NH₃), a polar aprotic solvent such as propanone or ethanol, and a good leaving group (I⁻ > Br⁻ > Cl⁻ > F⁻).

A typical exam task is to draw the mechanism for the hydrolysis of bromoethane with aqueous NaOH, showing the curly arrow from the lone pair on the hydroxide oxygen to the δ⁺ carbon and a second arrow from the C–Br bond to the bromine. The product is ethanol and bromide ions. Remember that the hydroxide ion is regenerated when the alcohol is a catalyst, but in the hydrolysis with aqueous alkali the OH⁻ is consumed as a reagent.

4. Nucleophilic Substitution: The SN1 Mechanism | 亲核取代:SN1 机理

SN1 stands for substitution, nucleophilic, unimolecular. It proceeds in two steps: first the C–X bond breaks heterolytically to give a carbocation intermediate, then the nucleophile attacks the carbocation. The first step is slow and rate-determining, so the rate depends only on the halogenoalkane:

rate = k[(CH₃)₃CBr]

Carbocation stability follows the order tertiary > secondary > primary > methyl, because alkyl groups release electron density inductively and by hyperconjugation. This explains why tertiary halogenoalkanes react fastest by SN1. The carbocation is planar, so the nucleophile can attack from either face with equal probability; when the carbon is chiral, the product is a racemic mixture of two enantiomers, showing optical inactivity.

SN1 is favoured by a polar protic solvent (water, ethanol) that stabilises both the carbocation and the halide ion, a weak nucleophile, and a tertiary substrate. In CIE examinations, you should be able to explain the difference between SN1 and SN2 in terms of rate equations, stereochemistry, and the effect of changing the halogenoalkane from primary to tertiary. A useful comparison table is given below.

Feature SN1 SN2
Steps Two One (concerted)
Rate equation k[RX] k[RX][Nu⁻]
Stereochemistry Racemisation Inversion
Best substrate Tertiary Primary

5. Elimination Reactions |

Halogenoalkanes can also lose HX to form alkenes. In the E2 mechanism, a base removes a proton from the carbon adjacent to the one bearing the halogen while the C–X bond breaks, giving the alkene in one step. Hot, concentrated KOH in ethanol (a non-aqueous solvent) favours elimination, whereas dilute aqueous KOH favours substitution. The base attacks the β-hydrogen, so the two groups must be able to adopt an anti-periplanar arrangement.

Elimination usually follows Zaitsev' stabilised by more alkyl groups. However, with bulky bases or with a poor leaving group, the less substituted alkene may predominate (the Hofmann product). Tertiary halogenoalkanes can also eliminate by an E1 route that shares the carbocation intermediate with SN1, so the two products often appear together.

When predicting the outcome of a reaction between a halogenoalkane and a base, ask three questions: is the base strong and bulky, is the solvent protic or aprotic, and is the temperature high? High temperature and strongly basic conditions push the reaction towards elimination; mild, aqueous and cooler conditions push it towards substitution. Examiners frequently test this competition by asking you to explain why a change of solvent or temperature changes the major product.

6. Electrophilic Addition to Alkenes |

The π bond of an alkene is electron-rich and acts as a nucleophile. In electrophilic addition, an electrophile attacks the π bond, a carbocation forms, and a nucleophile then attacks the carbocation. For example, ethene reacts with HBr to give bromoethane, and with steam in the presence of a phosphoric acid catalyst at about 300 °C and 60 atm to give ethanol.

is 2-bromopropane rather than 1-bromopropane. This observation is the basis of Markovnikov' colourised, from orange-brown to colourless, and this is the standard test for a C=C bond.

aturated polymer such as poly(ethene) or poly(chloroethene). In these reactions the π bond opens and no small molecule is eliminated, so addition polymerisation differs fundamentally from condensation polymerisation, where water or another small molecule is lost.

7. Electrophilic Substitution in Arenes |

Benzene is unusually stable because its six π electrons are delocalised over the ring. Rather than adding across the ring and losing that delocalisation, benzene substitutes a hydrogen and keeps the aromatic system intact. The general mechanism involves an electrophile attacking the ring to form a delocalised but positively charged intermediate (the arenium ion), followed by loss of H⁺ to restore the aromatic ring.

Nitration uses a mixture of concentrated nitric acid and concentrated sulfuric acid at 50–60 °C. The sulfuric acid protonates the nitric acid, which then loses water to form the nitronium ion NO₂⁺, the true electrophile. Halogenation requires a halogen carrier catalyst such as AlCl₃ or FeBr₃, which polarises the halogen molecule to generate a stronger electrophile. Both reactions give monosubstituted products if the temperature is controlled.

Friedel–Crafts reactions introduce alkyl or acyl groups: an alkyl halide with anhydrous AlCl₃ gives an alkylbenzene, and an acyl chloride with AlCl₃ gives a ketone. Acylations are generally more useful because the product is less reactive than the starting material, so polysubstitution is avoided. Substituents already on the ring direct incoming electrophiles: –OH, –NH₂ and alkyl groups are 2,4-directing and activating, while –NO₂, –COOH and –SO₃H are 3-directing and deactivating.

8. Nucleophilic Addition to Carbonyl Compounds |

s more electronegative than carbon, leaving the carbon δ⁺ and therefore vulnerable to nucleophiles. In nucleophilic addition, the nucleophile attacks the carbonyl carbon, the π bond breaks, and the oxygen becomes negatively charged; protonation then gives the final product. This differs from alkene chemistry, where the electron-rich π bond is attacked by an electrophile.

C=O 键是极性键,因为氧的电负性大于碳,使碳显 δ⁺,因而易受亲核试剂进攻。在亲核加成中,亲核试剂进攻羰基碳,π 键断裂,氧带负电;随后质子化得到最终产物。这与烯烃化学不同:烯烃中富电子的 π 键是被亲电试剂进攻的。

A classic example is the reaction of aldehydes and ketones with hydrogen cyanide, catalysed by a small amount of KCN. The cyanide ion attacks the carbonyl carbon to give a hydroxynitrile (cyanohydrin), which contains a new chiral centre. Reduction with NaBH₄ in aqueous ethanol or with LiAlH₄ in dry ether gives a primary alcohol from an aldehyde and a secondary alcohol from a ketone.

Reactivities differ: aldehydes are more reactive than ketones towards nucleophiles because ketones have two electron-releasing
