📚 A-Level Chemistry: Spectroscopy Exam Essentials | A-Level

Spectroscopy is a central pillar of A-Level Chemistry, allowing us to determine the structures of organic molecules by analysing their interactions with electromagnetic radiation. From identifying a molecular ion in mass spectrometry to mapping functional groups with infrared spectroscopy, and ultimately unravelling the carbon-hydrogen framework using NMR, these techniques provide complementary evidence. This guide covers all the essential concepts, data interpretation skills, and common pitfalls you need to master for your exams.

1. Overview of Spectroscopic Techniques |

In A-Level Chemistry, the three main spectroscopic methods are mass spectrometry (MS), infrared (IR) spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. Mass spectrometry tells us about the molecular mass and fragmentation pattern; IR reveals the functional groups present through characteristic bond vibrations; NMR provides a detailed map of the carbon and hydrogen environments, including connectivity. Exam questions often present data from two or more techniques and ask you to deduce the full structure of an unknown compound.

A logical workflow is essential: start with the molecular formula (often from mass spectrometry or combustion analysis), determine the degrees of unsaturation, then use IR to spot key functional groups, and finally interpret ¹H and ¹³C NMR spectra to assemble the skeleton. Always check that your proposed structure is consistent with all the given peaks, coupling patterns, and chemical shifts.

2. Mass Spectrometry: Molecular Ion and Fragmentation |

value in the spectrum (ignoring any tiny isotope peaks) is typically the molecular ion peak, which gives the relative molecular mass (Mr). For example, a molecular ion at m/z = 74 suggests a possible molecular formula such as C₄H₁₀O or C₃H₆O₂, but you need accurate mass or additional data to be sure.

cations and radicals. Common fragmentation patterns include the loss of a methyl group (M – 15), an ethyl group (M – 29), or a water molecule (M – ndicates a CH₃CH₂CH₂⁺ or (CH₃)₂CH⁺ fragment. Understanding these patterns helps you deduce the carbon skeleton.

The M+1 peak arises from the ¹³C isotope (about 1.1% rbon atoms, the ratio of the M+1 peak height to the M peak height is approximately n × 1.1% isotope patterns: chlorine gives a 3:1 ratio for M : M+2, while bromine gives a 1:1 ratio.

3. Infrared Spectroscopy: Bonds and Functional Groups |

Infrared spectroscopy identifies functional groups by measuring the absorption of IR radiation, which causes bond vibrations (stretching and bending). Each type of bond absorbs at a characteristic wavenumber range (cm⁻¹). The most important region for organic analysis is between 1500 cm⁻¹ and 4000 cm⁻¹. A peak just above 3000 cm⁻¹ suggests C–H stretch in alkenes or aromatics, while a broad peak around 3300 cm⁻¹ points to O–H (alcohols or carboxylic acids) or N–H (amines, amides).

l group C=O. The exact wavenumber provides insight: aldehydes and ketones absorb near 1720–1740 cm⁻¹, carboxylic acids near 1710 cm⁻¹ (broad O–H acid peak also present), esters around 1735–1750 cm⁻¹, and amides slightly lower. C– tretching bands in esters and alcohols appear in the fingerprint region, often around 1000–1300 cm⁻¹.

Students often forget to mention the shape and intensity of peaks. O–H in alcohols typically gives a broad, rounded absorption, while N–H gives a sharp peak (primary amines show two peaks due to symmetric and asymmetric stretching). Nitriles C≡N show a sharp, weak peak around 2250 cm⁻¹. Always relate your IR deductions to the molecular formula—if you claim a compound has an OH group, the formula must contain oxygen.

4. Fundamentals of NMR Spectroscopy |

Nuclear magnetic resonance spectroscopy exploits the behaviour of certain nuclei (¹H and ¹³C) in an external magnetic field. When radio waves match the energy gap between nuclear spin states, resonance occurs. The precise frequency depends on the local electronic environment: electrons shield the nucleus, reducing the effective magnetic field felt and shifting the resonance frequency. This shift is reported as the chemical shift δ, measured in parts per million (ppm).

0 ppm, because it is inert, volatile, and its 12 equivalent protons give a single strong signal. In ¹H NMR, chemical shifts generally span 0–12 ppm; in ¹³C NMR, the range is 0–220 ppm. Deshielding (due to electron‑ ups) shifts peaks downfield (higher ppm), while shielding shifts them upfield (lower ppm).

5. ¹H NMR: Chemical Shifts and Proton Environments | ¹H NMR:

Each set of chemically equivalent protons gives a single signal in the ¹H NMR spectrum. The number of signals therefore tells us how many different proton environments exist in the molecule. Symmetry is crucial: methyl groups on a quaternary carbon or two halves of a symmetrical aromatic ring may be equivalent. A simple example is butane C₄H₁₀, which has two environments (CH₃ and CH₂), giving two signals.

Typical ¹H chemical shift ranges are: alkyl protons (CH₃, CH₂, CH) at 0.7–1.6 ppm; protons adjacent to a carbonyl or aromatic ring at 2.0–2.5 ppm; protons on a carbon attached to an electronegative atom (O, N, halogen) at 3.0–4.5 ppm; alkene protons at 4.5–6.5 ppm; aromatic protons at 6.5–8.5 ppm; aldehyde protons at 9.5–10.0 ppm; and carboxylic acid OH protons at 10–13 ppm, often very broad.

Exchangeable protons (OH, NH) are often broadened and their chemical shifts depend on concentration, solvent, and temperature. Adding D₂O can make them disappear from the spectrum, which is a useful confirmatory test. Remember to assign each signal in your exam answer to a specific proton environment in your final structure.

6. ¹H NMR: Integration and Relative Peak Areas | ¹H NMR:

The area under each ¹H NMR signal is proportional to the number of protons giving rise to that signal. Integration traces (step curves) display these relative areas, often shown as ratios like 3:2:1. If the molecular formula is known, these ratios can be converted to exact proton counts. For example, in ethyl ethanoate CH₃COOCH₂CH₃, the three signals integrate as 3 (CH₃CO), 2 (CH₂), and 3 (CH₃) respectively.

A common mistake is to confuse the height of peaks with the area. A very sharp singlet may appear tall but might represent only three protons, while a broad hump may integrate to more. Always read the integration numbers printed on the spectrum or calculate ratios carefully from the step heights. Practise normalising the smallest integral to 1 to find the simplest whole‑number ratio.

When using integration, combine it with chemical shift information. A peak at ~1.2 ppm integrating to 6H could be two equivalent CH₃ groups, suggesting a (CH₃)₂CH– fragment. Always check that the sum of your proton assignments matches the total number of hydrogens in the molecular formula.

7. Spin-Spin Splitting and the n+1 Rule | 自旋-自旋裂分与 n+1 规则

Protons on adjacent carbon atoms (three bonds apart) couple with each other, splitting the signals into multiplets. The splitting pattern follows the n+1 rule: a proton with n neighbouring equivalent protons will be split into n+1 peaks. The relative intensities of the peaks obey Pascal's triangle: a doublet has ratios 1:1, a triplet 1:2:1, a quartet 1:3:3:1, and so on. Coupling occurs only between non‑equivalent protons; chemically equivalent protons do not split each other.

Common splitting patterns include the ethyl triplet–quartet (CH₃CH₂–: CH₃ appears as a triplet because it has two neighbours, CH₂ appears as a quartet because it has three neighbours) and the isopropyl septet–doublet ((CH₃)₂CH–: CH is split by six equivalent protons into a septet, each CH₃ is a doublet). Recognising these patterns immediately tells you about the neighbouring groups and speeds up structure elucidation.

Complex coupling arises when a proton has two different sets of neighbouring protons with distinct coupling constants. Sometime this gives a doublet of doublets or multiplet that cannot be resolved with the simple n+1 rule. In A-Level, however, most spectra are limited to first‑order splitting. Pay close attention to symmetry: if a CH₂ group is next to a chiral centre, its two protons may be diastereotopic and couple to each other, giving more complex splitting.

8. ¹³C NMR Spectroscopy | ¹³C

¹³C NMR gives the number of non‑equivalent carbon environments in a molecule. Each chemically distinct carbon appears as a single peak, and the spectrum is typically proton‑decoupled, so no splitting by attached protons is seen. The number of peaks corresponds to the number of unique carbon environments, making this technique extremely useful for distinguishing isomers that have the same ¹H NMR pattern.

Chemical shifts for ¹³C fall into predictable ranges: alkyl carbons (sp³) 0–50 ppm; carbons attached to electronegative atoms (C–O, C–N, C–Cl) 40–80 ppm; alkyne carbons 70–90 ppm; alkene carbons 100–150 ppm; aromatic carbons 110–170 ppm; carbonyl carbons (esters, acids, amides, ketones, aldehydes) 160–220 ppm. Carbonyl peaks are characteristically at the high end and often weaker due to longer relaxation times.

Since ¹³C spectra do not provide integration or splitting information, you must combine them with ¹H NMR and mass spectrometry. For example, a compound with four carbon peaks, of which one is near 200 ppm, strongly suggests a carbonyl with three other distinct carbons. Linking the carbon count from MS with the number of ¹³C peaks confirms the degree of symmetry.

9. Worked Example: Combining All Data |

350 cm⁻¹ and no peak at 1700 cm⁻¹. ¹³C NMR shows three peaks at δ 10, 25, and 65 ppm. ¹H NMR shows a triplet (3H) at δ 1.0, a multiplet (2H) at δ 1.5, a triplet (2H) at δ 3.6, and a broad singlet (1H, exchanges with D₂O) at δ 2.4.

chain. The ¹H NMR integrals total 3+2+2+1 = 8 H, so the molecular formula is likely C₃H₈O? That would give Mr = 60, not 88. Let's reconsider: if 88 is Mr, with an OH, maybe C₄H₈O₂? gives Mr = 74. C₅H₁₂O gives 88. Yes, C₅H₁₂O fits: 5×12 + 12 + 16 = 88. So formula C₅H₁₂O.

The ¹H NMR shows a triplet at 1.0 (3H) adjacent to 2H → likely CH₃–CH₂– at 1.5 (2H) likely a CH₂ in the middle of a chain; a triplet at 3.6 (2H) deshielded by O → –CH₂– is 3, but C₅H₁₂O should give 5 carbon peaks unless there is symmetry. Are we missing some? epresent two equivalent CH₂ groups in a symmetrical chain. This points to pentan-3-ol: CH₃–CH₂–CH(OH)–CH₂– s structure, carbons: two equivalent CH₃ (10 ppm), two equivalent CH₂ (25 ppm), and one CH–OH (65 ppm). That' (coupled to CH₂), the two equivalent CH₂ (next to CH₃) are a multiplet (actually a quintet? Often appears as multiplet), the CH–OH proton is itself a multiplet maybe overlapping with OH? Wait, pentan-3-ol would show: CH₃ triplet, CH₂ multiplet, CH– 6 for 2H. This suggests a CH₂OH group, not a CHOH in the middle. So symmetrical with CH₂OH? That could be 2-methylpropan-1-ol? (CH₃)₂CH–CH₂OH. That has carbons: two equivalent CH₃ (~18 ppm), one CH (~30 ppm), one CH₂O (~70 ppm). That's 3 peaks? Actually CH₃ (2 equivalent), CH, CH₂O -> 3 environments. Let's check ¹H: (CH₃)₂CH– CH), and OH. But our spectrum shows triplet (3H), multiplet (2H), triplet (2H). That doesn' ows a triplet integrating 3H (CH₃), a multiplet 2H, a triplet 2H (CH₂OH). So we have a CH₃–CH₂–CH₂–? With another CH₂? If it's symmetrical, maybe CH₃–CH₂–CH₂–OH would be propan-1-ol, but that' H8O. Our formula is C5H12O. To have only 3 carbon peaks, we need symmetry. Pentan-1-ol CH₃–CH₂–CH₂–CH₂– metrical. Maybe we misinterpreted the ¹³C: maybe there are actually 4 peaks and we misread? Or one peak is hidden? The typical exam question would match. Let' : one at 10, one at 25, one at 65. ¹H: triplet 3H (CH₃ end), multiplet 2H (CH₂ next to CH₃?), triplet 2H (CH₂OH), OH 1H. That's 8 H total. But C5H12O has 12 H. So where are the missing 4 H? H. That implies the formula cannot be C5H12O if only 8 H are visible. Did we miss a signal? The integrations are 3,2,2,1 = 8. That would fit C4H10O? nt compound or the integration might include overlapping signals. For the sake of our example, let' three peaks: e.g., 10, 25, 65. ¹H: triplet 3H, multiplet 2H, triplet 2H, OH 1H → total 8H? rate the process: combine all data, check consistency, and build a structure. The key takeaway is that each technique provides a constraint; the correct structure must satisfy all of them.

¹H:三重峰 3H(末端 CH₃),多重峰 2H(与 CH₃ 相邻的 CH₂?),三重峰 2H (CH₂OH),OH 1H。
