1. Overview of Photosynthesis |
Photosynthesis is the process by which photoautotrophs, such as plants, algae, and cyanobacteria, capture light energy and use it to convert carbon dioxide and water into glucose and oxygen. The overall balanced equation is:
6CO₂ + 6H₂O → C₆H₁₂O₆ + 6O₂
This seemingly simple summary hides two distinct sets of reactions: the light-dependent reactions that trap light energy and split water, and the light-independent reactions (Calvin cycle) that use that trapped energy to fix carbon. The two stages are linked by the energy carriers ATP and reduced NADP (NADPH).
2. Chloroplast Structure and Adaptations |
In eukaryotic plant cells, photosynthesis takes place inside chloroplasts. The key structural features are adapted for maximal efficiency:
- Thylakoids: flattened membrane sacs stacked into grana; the site of the light-dependent reactions. Their large surface area maximises light absorption and houses photosystems, electron carriers, and ATP synthase.
- Grana: stacks of thylakoids that increase surface area and allow close packing of photosystems and proton gradients.
- Stroma: the fluid-filled matrix surrounding thylakoids containing enzymes for the Calvin cycle, RuBP, DNA, and ribosomes. It has a suitable pH and ion concentration for carbon fixation.
- Inner and outer membranes: the inner membrane encloses the stroma, while the outer membrane is permeable to small molecules. The thylakoid membrane is impermeable to H⁺, allowing a proton gradient to be built up.
3. Photosynthetic Pigments and Light Absorption |
Chloroplasts contain several pigments that absorb light of specific wavelengths. The two main groups are chlorophylls and carotenoids. Chlorophyll a is the primary pigment, directly involved in the light reactions; chlorophyll b, xanthophylls, and carotenoids are accessory pigments that broaden the spectrum of light that can be utilised.
The absorption spectrum shows which wavelengths are absorbed by each pigment. The action spectrum shows the rate of photosynthesis at different wavelengths. The close match between the action spectrum and the combined absorption spectrum of all pigments demonstrates that these pigments are responsible for driving photosynthesis. Chlorophylls absorb mainly blue and red light, reflecting green, which is why leaves appear green.
Carotenoids protect chlorophyll from photo-oxidation by absorbing excess energy and dissipating it as heat, especially under high light intensity.
4. The Light-Dependent Reactions in Detail |
The light-dependent reactions take place in the thylakoid membrane and involve two photosystems: photosystem II (PSII) and photosystem I (PSI), linked by an electron transport chain. The key events are photolysis of water, electron transfer, and chemiosmosis.
1. A photon strikes PSII, exciting an electron in chlorophyll a to a higher energy level. This electron is passed to the primary electron acceptor, then moves along the electron transport chain (plastoquinone, cytochrome b6f complex, plastocyanin) to PSI. The energy released during these redox reactions pumps H⁺ from the stroma into the thylakoid lumen, building up a proton gradient.
H⁺ + 4e⁻ + O₂. This occurs in the thylakoid lumen, releasing oxygen as a by-product. The protons contribute further to the gradient.
3. Meanwhile, a photon also excites an electron in PSI, which is passed to another acceptor and eventually to ferredoxin. The enzyme NADP⁺ reductase then catalyses the reduction of NADP⁺ to NADPH using these electrons and H⁺ from the stroma.
The proton gradient drives ATP synthesis via ATP synthase (chemiosmosis). H⁺ ions flow back into the stroma through this enzyme, coupling the flow to the phosphorylation of ADP to ATP. This entire linear path is called non-cyclic photophosphorylation.
5. Photophosphorylation: Cyclic and Non-Cyclic |
In non-cyclic photophosphorylation (described above), both NADPH and ATP are produced, water is split, and oxygen is released. Electrons from PSI are used to reduce NADP⁺, so they do not return to PSI.
In cyclic photophosphorylation, only PSI is involved. The electron excited from PSI is passed to ferredoxin but then returned to the cytochrome b6f complex and back to PSI via plastocyanin. This electron cycling pumps H⁺ across the membrane, producing a proton gradient that drives ATP synthesis, but no NADPH is formed and no water is split. This process provides extra ATP to meet the high demands of the Calvin cycle, which uses more ATP than NADPH.
6. The Calvin Cycle: Carbon Fixation |
The Calvin cycle occurs in the stroma and uses the ATP and NADPH from the light reactions to fix CO₂ into organic molecules. It consists of three main phases: carbon fixation, reduction, and regeneration of the CO₂ acceptor, ribulose bisphosphate (RuBP).
1. Carbon fixation: CO₂ combines with RuBP (5-carbon sugar) in a reaction catalysed by RuBisCO, forming an unstable 6-carbon intermediate that immediately splits into two molecules of 3-phosphoglycerate (PGA), a 3-carbon acid.
2. Reduction: Each PGA is phosphorylated by ATP (forming 1,3-bisphosphoglycerate) and then reduced by NADPH + H⁺ to form glyceraldehyde-3-phosphate (G3P), a triose phosphate. For every 3 CO₂, 6 G3P are produced. One G3P exits the cycle to form glucose, sucrose, starch, or other carbohydrates, while the remaining 5 G3P are used to regenerate RuBP.
3. Regeneration: Using additional ATP, the 5 G3P molecules undergo a series of rearrangements to regenerate 3 molecules of RuBP (5C each), enabling the cycle to continue. The net consumption for one G3P is 9 ATP and 6 NADPH (for 3 CO₂).
7. C3, C4, and CAM Plants | C3、C4 与 CAM 植物
The basic C3 pathway (Calvin cycle) is common to all plants, but some have evolved adaptations to reduce photorespiration – the wasteful process in which RuBisCO fixes O₂ instead of CO₂, leading to loss of fixed carbon. Photorespiration is significant in hot, dry conditions when stomata close and CO₂ concentration drops inside the leaf.
| Feature | C3 Plants | C4 Plants | CAM Plants |
|---|---|---|---|
| Initial CO₂ fixation | PGA (3C) by RuBisCO in mesophyll cells | Oxaloacetate (4C) by PEP carboxylase in mesophyll; then malate to bundle sheath | Oxaloacetate (4C) at night; malate stored in vacuole; CO₂ released during day |
| Site of Calvin cycle | Mesophyll cells | Bundle sheath cells (Kranz anatomy) | Mesophyll cells (temporal separation) |
| Photorespiration | High in hot/dry | Minimal | Minimal |
| Examples | Rice, wheat, soybean | Maize, sugarcane, sorghum | Cacti, pineapples, succulents |
In C4 plants, CO₂ is concentrated around RuBisCO in bundle sheath cells, so the enzyme is saturated with CO₂ and photorespiration is suppressed. The C4 pathway consumes more ATP but is advantageous under high light and temperature.
CAM plants fix CO₂ into malate at night (stomata open) and release it for the Calvin cycle during the day (stomata closed). This temporal separation minimises water loss and photorespiration.
8. Limiting Factors of Photosynthesis |
The rate of photosynthesis is controlled by the factor that is in shortest supply – the limiting factor. The main ones are light intensity, carbon dioxide concentration, and temperature. At low light intensity, the rate increases linearly but eventually levels off as another factor (e.g., CO₂) becomes limiting. CO₂ concentration shows a similar saturation curve. Temperature affects enzyme activity, especially RuBisCO: as temperature rises, kinetic energy and reaction rate increase, but beyond an optimum (around 25–30°C for many C3 plants), enzymes begin to denature, and photorespiration increases.
Water availability indirectly limits photosynthesis because lack of water causes stomatal closure, reducing CO₂ entry and increasing photorespiration. The principle of limiting factors is often illustrated using the Blackman curve or by experiments with Elodea counting oxygen bubbles at varying distances from a light source, or by measuring the uptake of CO₂ or production of O₂ using sensors or chloroplast suspensions with redox indicators (e.g., DCPIP).
9. Measuring Photosynthesis: Experimental Techniques |
Several classic experiments help explore photosynthetic rate and pigment separation:
a) Oxygen production by aquatic plants (Elodea): Place a shoot of Elodea in water with sodium hydrogencarbonate (supplies CO₂), expose to light, and count bubbles or measure volume of oxygen produced over time. Vary light intensity (lamp distance), CO₂ concentration, or temperature. Use a light filter to investigate wavelength effects.
b) DCPIP colour-change assay with isolated chloroplasts: Isolate chloroplasts from spinach leaves. In the light-dependent reactions, electrons are transferred to DCPIP (an artificial electron acceptor) instead of NADP⁺. DCPIP is blue when oxidised and becomes colourless when reduced. The rate of decolourisation indicates the rate of electron transport, thus the rate of the light reactions. Add DCPIP to chloroplast suspension and measure absorbance decrease at 600 nm.
c) Paper chromatography of photosynthetic pigments: mixture. As the solvent rises, pigments separate based on their solubility and affinity for the stationary phase. Identify pigments by their colour and Rf values: carotene (top, yellow-orange), pheophytin (grey), chlorophyll a (blue-green), chlorophyll b (yellow-green), xanthophylls (yellow). Rf = distance moved by pigment ÷ distance moved by solvent.
10. Key Equations and Energy Conversions |
It is vital to recall the precise stoichiometry and energetic relationships in the CIE exam:
- Photolysis: 2H₂O → 4H⁺ + 4e⁻ + O₂
- NADP⁺ reduction: NADP⁺ + 2H⁺ + 2e⁻ → NADPH + H⁺
- ATP synthesis: ADP + Pᵢ → ATP (catalysed by ATP synthase)
The Calvin cycle produces one molecule of glucose from 6 CO₂, using 18 ATP and 12 NADPH. The overall equation for glucose synthesis from the combined stages can be written as:
6CO₂ + 12H₂O + 18ATP + 12NADPH + 12H⁺ → C₆H₁₂O₆ + 6O₂ + 18ADP + 18Pᵢ + 12NADP⁺
Energy conversions: light energy is absorbed by pigments and converted into chemical energy in the form of ATP and NADPH (short-term energy carriers). These are then used to synthesise stable organic molecules such as glucose, which store chemical potential energy. The thylakoid proton gradient is a form of potential energy converted into kinetic energy during H⁺ efflux, driving chemical energy in ATP. Light energy is thus ultimately transferred into chemical bond energy.
