1. Overview of Photosynthesis |
₂O₆ + 6O₂. Water is oxidised to oxygen, and carbon dioxide is reduced to carbohydrate. The process consists of two main stages: the light-dependent reactions, which capture light energy and produce ATP and reduced NADP, and the light-independent reactions (Calvin cycle), which use these products to fix CO₂ and synthesise glucose.
2. Chloroplast Structure |
Chloroplasts are surrounded by a double membrane. Inside, the stroma contains enzymes for the Calvin cycle, starch grains and lipid droplets. A network of flattened membrane sacs called thylakoids is stacked into grana. Thylakoid membranes house photosynthetic pigments, electron carriers and ATP synthase. The arrangement of grana connected by stromal lamellae provides a large surface area for light absorption and efficient energy transfer. The compartmentalisation of the thylakoid space and stroma is crucial for chemiosmosis.
3. Photosynthetic Pigments & Spectra |
Primary pigment chlorophyll a and accessory pigments (chlorophyll b, carotenoids) are arranged in light-harvesting complexes. Each pigment absorbs specific wavelengths of light. The absorption spectrum shows the wavelengths absorbed by a pigment, while the action spectrum illustrates the rate of photosynthesis at different wavelengths. The overlap between the two confirms that absorbed light drives photosynthesis. Accessory pigments broaden the range of usable wavelengths and protect against photo-oxidation.
4. Light-Dependent Reactions: Non-Cyclic Photophosphorylation |
then passed along an electron transport chain. Photolysis of water (2H₂O → 4H⁺ + 4e⁻ + O₂) replaces these electrons, releasing oxygen. As electrons move, H⁺ ions are pumped into the thylakoid space, creating a proton gradient. ATP synthase uses this gradient to generate ATP (chemiosmosis). Meanwhile, PSI re-energises electrons and transfers them to NADP⁺, forming reduced NADP. Products: ATP, reduced NADP and O₂.
非循环光合磷酸化涉及光系统 II(PSII)和光系统 I(PSI)。
5. Light-Dependent Reactions: Cyclic Photophosphorylation |
Cyclic photophosphorylation involves PSI only. Excited electrons from PSI travel through an electron transport chain and return to PSI. This process generates a proton gradient for ATP synthesis but does not produce reduced NADP or oxygen. It allows the chloroplast to produce extra ATP when the Calvin cycle demands more ATP than reduced NADP.
|---|---|---|
| Photosystems 光系统 | PSII and PSI | PSI only |
| Electron source 电子来源 | Photolysis of water | PSI itself |
| End products 最终产物 | ATP, reduced NADP, O₂ | ATP only |
| O₂ produced? 释放 O₂? | Yes | No |
6. Light-Independent Reactions: The Calvin Cycle |
The Calvin cycle occurs in the stroma and uses ATP and reduced NADP from the light-dependent reactions. It has three main phases: carboxylation (carbon fixation), reduction, and regeneration. RuBP (ribulose bisphosphate) combines with CO₂, catalysed by RuBisCO, to form two molecules of GP (glycerate 3-phosphate). GP is reduced to TP (triose phosphate) using ATP and reduced NADP. Most TP is recycled to regenerate RuBP, while some is used to synthesise glucose, sucrose, amino acids and lipids. For every six CO₂ molecules fixed, one hexose sugar can be formed.
RuBP(核酮糖二磷酸)在 RuBisCO 催化下与 CO₂ 结合,生成两分子 GP(甘油酸‑3‑磷酸)。
CO₂, ATP, reduced NADP
7. Factors Limiting Photosynthesis |
At any given moment, the rate of photosynthesis is limited by the factor in shortest supply – the law of limiting factors. Light intensity, CO₂ concentration and temperature are the main abiotic factors. At low light intensity, the light-dependent reactions are restricted. When CO₂ concentration is low, RuBisCO cannot fix carbon efficiently. Temperature affects enzyme activity; at low temperatures, kinetic energy is reduced, while at high temperatures RuBisCO can catalyse photorespiration and enzymes may denature. Agricultural practices often manipulate these factors to optimise crop yield.
C₄ plants (e.g. maize, sugarcane) have evolved a spatial separation of initial CO₂ fixation and the Calvin cycle. Their leaves exhibit Kranz anatomy: bundle sheath cells tightly surround vascular bundles, and mesophyll cells are arranged around them. CO₂ is first fixed in mesophyll cells by PEP carboxylase into a 4‑carbon compound (oxaloacetate). This compound is transported to bundle sheath cells, where CO₂ is released and enters the Calvin cycle. PEP carboxylase has a high affinity for CO₂ and does not fix oxygen, virtually eliminating photorespiration. This adaptation increases photosynthetic efficiency in hot, dry climates.
Crassulacean Acid Metabolism (CAM) plants (e.g. cacti, succulents) display a temporal separation of CO₂ uptake and fixation. Their stomata open at night to fix CO₂ into malate, which is stored in vacuoles. During the day, stomata close to conserve water, and malate is decarboxylated to release CO₂ for the Calvin cycle. This arrangement allows CAM plants to thrive in arid environments by minimising water loss while still performing photosynthesis.
