This article provides a comprehensive breakdown of the key concepts you need to excel.
1. Neurone Structure and Types |
A neurone is a specialised cell adapted to transmit electrical impulses. The three main types in a reflex arc are sensory neurones, relay neurones (interneurones), and motor neurones. Sensory neurones carry impulses from receptors to the central nervous system (CNS); they have a long dendron and a shorter axon. Relay neurones, found entirely within the CNS, connect sensory and motor neurones and have many short dendrites. Motor neurones transmit impulses from the CNS to effectors such as muscles or glands; they possess a long axon and short dendrites. All neurones share common features: a cell body containing the nucleus and organelles, dendrites that receive signals, and an axon that conducts impulses away from the cell body. Some axons are wrapped in a myelin sheath formed by Schwann cells, which insulates and speeds up conduction.
2. Establishment of the Resting Potential |
When a neurone is not transmitting an impulse, the inside of the axon is negatively charged relative to the outside; this is the resting potential, typically around –70 mV. The resting potential is created by the unequal distribution of ions across the membrane and the selective permeability of the membrane. Sodium-potassium pumps actively transport 3 Na⁺ out for every 2 K⁺ pumped in, using ATP. This creates a concentration gradient: Na⁺ concentration is higher outside, K⁺ concentration is higher inside. In the resting state, most voltage-gated sodium channels are closed, but some potassium channels are open, allowing K⁺ to leak out. The movement of K⁺ out of the cell down its concentration gradient makes the inside more negative. The large, negatively charged organic anions inside the cell also contribute. The equilibrium established is the resting potential.
3. Generation of an Action Potential |
An action potential is a rapid, temporary reversal of the membrane potential. It occurs when a stimulus causes the membrane to depolarise past the threshold potential (about –55 mV). The process has several phases:
- Depolarisation: Voltage-gated Na⁺ channels open, Na⁺ floods into the axon down its electrochemical gradient, making the inside more positive (up to about +40 mV).
- Repolarisation: At the peak, Na⁺ channels inactivate and voltage-gated K⁺ channels open. K⁺ rushes out, restoring the negative charge inside.
- Hyperpolarisation: The K⁺ channels are slow to close, so the potential briefly becomes more negative than the resting potential (overshoot).
- Return to resting potential: The sodium‑potassium pump restores ion concentrations.
During and immediately after an action potential, the axon is first in an absolute refractory period (no new action potential possible) and then a relative refractory period (a stronger-than-normal stimulus is required). This ensures unidirectional propagation and limits impulse frequency.
4. Propagation of the Action Potential |
Once an action potential is generated at one point on the axon, it does not stay there; it triggers the next region to depolarise. The influx of Na⁺ at the active site causes local currents: positive charge spreads to the adjacent polarised region, depolarising it to threshold and opening voltage‑gated Na⁺ channels. This creates a new action potential ahead. Behind the active zone, the membrane is in the refractory period, preventing backward propagation. Thus, the impulse moves in one direction along the axon like a wave.
5. Saltatory Conduction and Myelination |
nvier, where ion channels are concentrated. Local currents jump from one node to the next, a process called saltatory conduction. This vastly increases the speed of impulse transmission compared to unmyelinated fibres of the same diameter. Myelination also saves energy because fewer ions need to be pumped back. In the Edexcel specification, you may need to interpret data comparing conduction velocity in myelinated and non-myelinated axons, and explain how axon diameter (wider = faster, due to lower internal resistance) and temperature (higher temperature = faster diffusion of ions) affect speed.
6. Synapse Structure |
A synapse is a junction between two neurones, or between a neurone and an effector. The gap is called the synaptic cleft (about 20 nm). The presynaptic neurone’s terminal knob (synaptic knob) contains many mitochondria and synaptic vesicles filled with neurotransmitter, such as acetylcholine. The postsynaptic membrane possesses specific receptor proteins and is associated with channels and enzymes like acetylcholinesterase. The structure ensures that signals are transferred chemically, allowing integration and control of information flow.
7. Synaptic Transmission Process |
When an action potential arrives at the synaptic knob, it opens voltage-gated Ca²⁺ channels. Ca²⁺ ions enter the knob, causing synaptic vesicles to fuse with the presynaptic membrane and release neurotransmitter into the cleft by exocytosis. The neurotransmitter diffuses across the cleft and binds to specific receptors on the postsynaptic membrane. This binding opens ligand-gated Na⁺ channels, allowing Na⁺ to enter the postsynaptic neurone. If the resulting depolarisation reaches threshold, a postsynaptic action potential is triggered. To stop the signal, the neurotransmitter is removed by enzymatic breakdown (e.g. acetylcholinesterase breaks down acetylcholine) or re-uptake into the presynaptic knob.
8. Postsynaptic Integration and Summation |
A single postsynaptic neurone receives inputs from many presynaptic neurones. The small depolarisations produced by excitatory neurotransmitters (EPSPs) or hyperpolarisations by inhibitory neurotransmitters (IPSPs) are not individually strong enough to fire an action potential. The postsynaptic membrane integrates these signals by summation:
- Temporal summation: one presynaptic neurone fires repeatedly in quick succession, so the EPSPs add up over time.
- Spatial summation: several presynaptic neurones fire at the same time, and their EPSPs combine to reach threshold.
If the combined depolarisation at the axon hillock reaches threshold, an action potential is initiated. Inhibitory synapses can reduce the net depolarisation, preventing firing. This summation is the basis of decision-making in the nervous system.
9. Organisation of the Nervous System |
The mammalian nervous system is divided into the central nervous system (CNS) – brain and spinal cord – and the peripheral nervous system (PNS) – all nerves outside the CNS. The PNS is further subdivided into the sensory (afferent) division, conveying impulses to the CNS, and the motor (efferent) division, carrying impulses from CNS to effectors. The motor division comprises the somatic nervous system (voluntary control of skeletal muscle) and the autonomic nervous system (involuntary control of smooth muscle, cardiac muscle and glands). The autonomic nervous system has two branches: the sympathetic system, which prepares the body for 'fight or flight' (increases heart rate, dilates pupils, inhibits digestion), and the parasympathetic system, which promotes 'rest and digest' (decreases heart rate, stimulates digestion).
10. Reflex Arc |
A reflex action is a rapid, automatic, involuntary response to a stimulus, protecting the body from harm. The pathway is a reflex arc. For example, in the spinal withdrawal reflex (touching a hot object):
- The thermoreceptor in the skin generates an impulse in a sensory neurone.
- The sensory neurone transmits the impulse to the spinal cord via the dorsal root.
- In the spinal cord, the sensory neurone synapses with a relay neurone.
- The relay neurone synapses with a motor neurone.
- The motor neurone carries the impulse out of the ventral root to the effector muscle (e.g. biceps), which contracts to withdraw the hand.
This pathway often bypasses the brain for speed, although interneurones also send impulses to the brain for conscious awareness. The reflex arc demonstrates a simple, coordinated pathway of the nervous system.
11. Neuromuscular Junction |
The neuromuscular junction is a specialised cholinergic synapse between a motor neurone and a skeletal muscle fibre. It works exactly like a synapse but with a larger postsynaptic membrane folded into junctional folds to increase surface area. When an action potential arrives, it releases acetylcholine (ACh). ACh binds to nicotinic receptors on the muscle fibre membrane (sarcolemma), opening Na⁺ channels and depolarising the membrane. This triggers a muscle action potential, which spreads along the sarcolemma and down T-tubules, leading to Ca²⁺ release from the sarcoplasmic reticulum and muscle contraction via the sliding filament model. The enzyme acetylcholinesterase rapidly breaks down ACh to stop stimulation.
12. Effects of Drugs on Synapses |
Many drugs act by altering synaptic transmission. In the context of the Edexcel specification, you need to be able to predict and explain drug effects. For example:
- Cocaine blocks the re-uptake of dopamine at synapses in the brain, prolonging the effect of dopamine, which overstimulates post-synaptic neurones and leads to feelings of euphoria.
- Nicotine mimics acetylcholine by binding to nicotinic receptors, causing depolarisation and producing stimulant effects.
- Organophosphates (some insecticides) inhibit acetylcholinesterase, so ACh accumulates in the cleft, leading to continuous stimulation of cholinergic synapses and neuromuscular junctions, causing muscle spasms and paralysis.
- Botulinum toxin prevents the release of ACh at neuromuscular junctions, causing muscle paralysis.
Understanding these mechanisms helps you analyse data on drug action and relate them to the normal function of synapses.
