📚 IGCSE AQA Biology: Cloning Revision Guide | IGCSE AQA
In the IGCSE AQA Biology syllabus, cloning is a core topic that explores how genetically identical organisms can be produced. Understanding both natural processes and artificial techniques, along with their benefits and drawbacks, is essential for exam success.
1. What is Cloning? |
Cloning is the production of genetically identical individuals. A clone carries exactly the same DNA as the parent organism. This means all the offspring produced by cloning are copies of one original individual. Cloning occurs naturally in many organisms, but humans can also carry out artificial cloning using various techniques.
In biology, even the asexual reproduction of bacteria by binary fission is a form of natural cloning. Many plants produce clones through runners or bulbs, and some animals, like identical twins, are natural clones. Artificial cloning uses laboratory methods to produce large numbers of identical organisms or embryonic cells.
2. Natural Cloning in Plants |
Many plants naturally reproduce asexually, generating clones without human intervention. This is a common adaptation that allows plants to spread rapidly in a suitable environment. Three key examples are runners, bulbs and tubers.
Runners (stolons): Strawberry plants send out horizontal stems called runners. These grow along the soil surface, and at their tips a new plantlet develops. Once the plantlet establishes roots, it becomes independent, yet it is genetically identical to the parent.
Bulbs: Onions and daffodils store food in underground leaf bases forming a bulb. New bulbs can split off from the parent bulb, each developing into a separate cloned plant.
Tubers: Potatoes produce swollen underground stems called tubers. Each tuber has 'eyes' (buds) that can grow into a new potato plant. All plants grown from tubers of the same potato plant are clones.
3. Artificial Cloning in Plants: Cuttings |
Taking cuttings is one of the simplest artificial cloning methods used by gardeners and commercial growers. A piece of stem or leaf is cut from a healthy parent plant. The cutting is placed into moist soil or water, where it produces new roots and grows into a new plant genetically identical to the donor.
To improve success rates, growers often dip the cut end into rooting hormone powder. This encourages faster root development. Cuttings are widely used to produce ornamental plants, fruit bushes and some trees. The advantage is that all new plants retain the desirable traits of the parent, such as flower colour or fruit quality.
4. Artificial Cloning in Plants: Tissue Culture |
Tissue culture (also called micropropagation) is a modern method that can produce thousands of identical plantlets from a tiny piece of plant tissue called an explant. The process is carried out in sterile conditions on a nutrient agar medium containing hormones.
The steps of tissue culture include: first, selecting a small piece of healthy tissue from the parent plant. The explant is sterilised to kill any microorganisms. It is then placed on a growth medium with nutrients and plant hormones (auxins and cytokinins). The cells divide to form a mass of undifferentiated cells called a callus. By adjusting hormone levels, the callus is induced to develop into tiny plantlets. Finally, these plantlets are transferred to soil to grow into full-sized plants.
A major benefit of tissue culture is that it produces virus‑free plants when meristem tissue is used. It also allows rapid propagation of rare or valuable plants in a small space.
5. Natural Cloning in Animals |
In animals, natural cloning is less common but does occur. The best-known example is identical twins. Identical twins form when a single fertilised egg (zygote) splits into two separate embryos very early in development. Because both embryos come from the same zygote, they share exactly the same genetic material and are natural clones of each other.
Some invertebrates, such as aphids, can reproduce asexually by parthenogenesis, where females produce offspring from unfertilised eggs. The young are clones of the mother. However, in the IGCSE exam, the focus is usually on identical twins as the key example of natural animal cloning.
6. Artificial Cloning in Animals: Embryo Transplants |
Embryo transplant cloning is used in agriculture to produce many genetically identical offspring from a single prized female animal. A cow or sheep with desirable traits (e.g., high milk yield) is given hormone treatment to produce multiple eggs. The eggs are fertilised in vitro using sperm from a high‑quality male. The resulting zygotes develop into early embryos.
When each embryo is a small ball of cells, it is split into several individual cells or smaller clumps. These are then implanted into surrogate mothers (ordinary females of the same species). The surrogate gives birth to offspring that are clones of the original embryo, not the surrogate. This technique rapidly multiplies desirable farm animals.
7. Artificial Cloning in Animals: Adult Cell Cloning |
Adult cell cloning, also known as somatic cell nuclear transfer (SCNT), allows scientists to create a clone using the nucleus from an adult body cell. The most famous example is Dolly the sheep, born in 1996 – the first mammal cloned from an adult somatic cell.
The process involves removing the nucleus from an unfertilised egg cell (an enucleated egg) and replacing it with the diploid nucleus from a body cell of the animal to be cloned. The egg is then stimulated to divide and develops into an embryo, which is implanted into a surrogate mother. The offspring is a clone of the original nucleus donor.
8. Steps of Somatic Cell Nuclear Transfer |
1. A mature unfertilised egg cell is collected from a female donor. The haploid nucleus of this egg cell is carefully removed using a micropipette, leaving an enucleated egg cell.
2. A diploid nucleus is taken from a body cell (e.g., a skin or udder cell) of the animal that is to be cloned. This provides the complete genetic blueprint of the desired individual.
3. The diploid nucleus is inserted into the enucleated egg cell. An electric shock is applied to fuse the nucleus with the egg cytoplasm and trigger cell division, mimicking the effect of fertilisation.
4. The fused cell begins dividing to form an embryo in laboratory culture. When the embryo reaches a suitable stage, it is implanted into the uterus of a surrogate mother.
5. The surrogate mother carries the pregnancy to term and gives birth. The newborn is a clone of the animal that provided the body cell nucleus, not of the egg donor or surrogate.
Because all the genetic information comes from the donated diploid nucleus, the resulting animal is genetically identical to the nucleus donor, effectively an adult cell clone.
9. Advantages of Cloning |
Cloning offers several significant benefits in agriculture, conservation and medicine. Genetically identical organisms with desirable characteristics can be produced reliably and quickly.
- Agricultural advantage: Farmers can multiply high‑yielding livestock or crop plants that are resistant to diseases, ensuring uniform product quality.
- Conservation: Endangered species can be cloned to increase population numbers, though care is needed to maintain genetic diversity.
- Medical research: The cloning of cells, tissues or organs (therapeutic cloning) may one day provide transplant material that is perfectly matched to the patient, reducing rejection risks.
- Uniform products: In horticulture, tissue culture guarantees that every plant is identical, which is important for the cut‑flower and ornamental plant industries.
10. Disadvantages and Ethical Issues |
Despite the benefits, cloning raises serious concerns. A major drawback is the lack of genetic variation. If all individuals are identical, a single disease or environmental change could wipe out the entire population. This is a significant risk for cloned crop plants and animals.
In animal cloning, many cloned embryos fail to develop, and clones are sometimes born with health problems or premature ageing. Dolly the sheep, for example, developed arthritis at an early age and died younger than normal. The low success rates and potential suffering raise animal welfare concerns.
Ethically, the possibility of human reproductive cloning is almost universally condemned, and it is illegal in most countries. There are also fears that ‘designer’ cloning could lead to devaluation of individual life. Moreover, the reduction of genetic diversity in agriculture could threaten food security if cloned monocultures become vulnerable to pests.
