1. What is Genetic Engineering? |
Genetic engineering, also called genetic modification (GM), is the direct manipulation of an organism's genome using biotechnology. It involves transferring a gene from one species into the DNA of another species, thereby giving the recipient organism a new, desirable characteristic.
The transferred gene is called the gene of interest. Once inserted and expressed, it leads to the production of a specific protein that was not originally made by the host organism. For example, a human gene can be put into a bacterium to make human insulin.
2. Basic Tools: Restriction Enzymes |
Restriction enzymes (restriction endonucleases) are proteins that cut DNA at specific sequences of bases, known as recognition sites. Each restriction enzyme recognises a particular sequence, usually 4–8 base pairs long, and cuts the DNA in a precise way.
Many restriction enzymes produce 'sticky ends' – short, single-stranded overhangs that can base-pair with complementary sticky ends on other DNA fragments cut by the same enzyme. This ability to create matching ends is vital for combining DNA from different sources.
For example, the restriction enzyme EcoRI recognises the sequence GAATTC and cuts between G and A, leaving sticky ends AATT. The sequence is palindromic, meaning it reads the same forwards and backwards on complementary strands.
DNA ligase is an enzyme that seals the sugar–phosphate backbone of DNA. After restriction enzymes cut DNA and fragments are joined by base pairing of sticky ends, ligase covalently bonds the fragments together, creating a continuous strand of DNA.
Without ligase, the recombinant DNA would remain only loosely held by hydrogen bonds and would not function stably inside a host cell. Ligase thus completes the construction of recombinant DNA molecules, such as the insertion of a gene into a plasmid.
4. Vectors: Plasmids and More | 载体:质粒及其他
A vector is a DNA molecule used to carry foreign genetic material into another cell. The most common vectors in IGCSE Edexcel biology are bacterial plasmids – small, circular DNA molecules found in bacteria, separate from the main chromosome.
To act as an effective vector, a plasmid must have: an origin of replication (so it can be copied inside the host), a multiple cloning site (a region containing many restriction enzyme recognition sites), and a selectable marker gene (often an antibiotic resistance gene) that allows scientists to identify cells that have taken up the plasmid.
Other vectors include viruses, but plasmids are the focus at IGCSE level because they are easy to manipulate and enter bacterial cells naturally through transformation.
Recombinant DNA (rDNA) is DNA that has been formed artificially by combining genetic material from two different sources. The process typically uses the same restriction enzyme to cut both the gene of interest from a donor genome and the plasmid vector.
This produces complementary sticky ends. The gene of interest and the cut plasmid are mixed together with DNA ligase, which covalently joins the sugar–phosphate backbones. The result is a recombinant plasmid carrying the foreign gene.
The recombinant plasmid is now ready to be introduced into a host cell, where it can multiply and express the inserted gene.
6. Inserting Recombinant DNA into Host Cells | 将重组DNA导入宿主细胞
The process of getting recombinant DNA into a host cell is called transformation (for bacteria) or transfection (for eukaryotic cells). For bacteria, cells are often treated with calcium chloride or subjected to heat shock, making their membranes permeable to plasmid uptake.
Once inside, the host cell begins to use the gene's instructions to synthesise the desired protein. If the plasmid contains an antibiotic resistance gene, scientists can grow the transformed cells on agar containing that antibiotic; only cells that have taken up the plasmid survive, allowing easy selection.
7. Example: Making Human Insulin |
One of the most important applications of genetic engineering is the production of human insulin by genetically modified bacteria. Before this technique, insulin was extracted from the pancreases of cattle or pigs, which could cause allergic reactions and was supply-limited.
The human insulin gene is isolated and inserted into a plasmid vector, then introduced into E. coli bacteria. The transformed bacteria are cultured in large fermenters, where they multiply and express the human insulin gene, producing large quantities of human insulin protein.
The insulin is then harvested, purified and used to treat people with diabetes. This method produces insulin identical to human insulin, reducing the risk of immune reactions and ensuring a consistent supply.
8. Genetically Modified (GM) Crops |
GM crops are plants whose DNA has been altered using genetic engineering techniques. The aim is usually to introduce a trait that does not occur naturally in that species, such as resistance to pests, tolerance to herbicides or improved nutritional content.
Common examples include Bt corn (which produces an insecticidal protein), herbicide-tolerant soybeans, and Golden Rice (engineered to produce beta-carotene, a precursor of vitamin A, to combat blindness in populations with vitamin A deficiency).
9. Herbicide-Resistant Crops |
Some GM crops have been engineered to be resistant to broad-spectrum herbicides, such as glyphosate. The gene inserted allows the plant to survive application of the herbicide, which kills all surrounding weeds.
This allows farmers to control weeds more easily, often with fewer applications and less soil disturbance. However, concerns exist about the development of herbicide-resistant 'superweeds' and the increased use of chemicals.
Insect-resistant GM crops contain a gene from the bacterium Bacillus thuringiensis (Bt). This gene codes for a protein (Bt toxin) that is poisonous to specific insect pests but harmless to humans and most other organisms.
When insects feed on the plant, the Bt toxin damages their digestive system, leading to death. This reduces the need for chemical insecticides, saving costs for farmers and reducing environmental impact from pesticide spraying.
However, there is a risk that insect populations may evolve resistance to Bt toxin over time, so refuge areas (non-GM crops planted nearby) are often used to delay this.
11. Benefits of Genetic Engineering |
Genetic engineering offers many potential benefits. In medicine, it enables the mass production of human proteins like insulin, growth hormone and clotting factors, which are purer and more effective than animal-derived versions.
In agriculture, GM crops can increase yields, reduce dependence on chemical pesticides, improve nutritional quality and allow crops to grow in harsh environments, such as drought-prone or saline soils. This may contribute to global food security.
Additionally, genetic engineering is used in research to study gene function, in gene therapy to treat genetic disorders, and in industry to produce enzymes and biofuels more sustainably.
12. Risks and Ethical Concerns |
Despite its advantages, genetic engineering raises several concerns. One risk is that inserted genes could transfer to other organisms, such as when GM crops cross-pollinate with wild relatives, potentially creating herbicide-resistant weeds or disrupting ecosystems.
Some people worry about food safety – whether GM foods might cause allergies or long-term health effects. Rigorous testing is required before GM foods are approved, but uncertainty remains for some consumers.
Ethically, there are debates about 'playing God' with nature, the patenting of life forms, and the control of GM technology by large corporations, which can disadvantage small farmers in developing countries. Informed, balanced discussion is therefore essential.
For IGCSE, you need to be able to evaluate both sides using scientific arguments, not simply personal opinion.
