📚 A-Level Biology: Proteins – Exam Essentials | A-Level
Proteins are one of the most diverse and essential macromolecules in living organisms. In A-Level Biology, mastering their structure – from amino acid sequence to functional quaternary assemblies – is fundamental to understanding processes such as enzyme action, cell signalling, and molecular transport. This revision guide walks through every examinable point: the chemistry of amino acids, the four levels of protein structure, the bonds that hold proteins together, denaturation, the Biuret test, and the structure-to-function links that examiners reward most often.
1. Amino Acids: The Building Blocks |
All proteins are polymers of amino acid monomers. A typical amino acid has a central α-carbon bonded to an amino group (–NH₂), a carboxyl group (–COOH), a hydrogen atom, and a variable R group (side chain). Because four different groups are attached to the same carbon, amino acids exist as optical isomers; only the L-isomers are used in protein synthesis.
0 standard amino acids, each with a unique R group that determines its chemical properties – y charged), or basic (positively charged). Glycine is the simplest amino acid, with R = H.
| Class of R group | R 基团类型 | Charge in solution | 溶液中的电荷 | Examples | 举例 |
|---|---|---|
Essential amino acids cannot be synthesised by the human body and must be obtained from the diet. In humans there are nine: histidine, isoleucine, leucine, lysine, methionine, phenylalanine, threonine, tryptophan and valine.
In aqueous solution, amino acids exist mainly as zwitterions: the amino group is protonated (–NH₃⁺) and the carboxyl group is deprotonated (–COO⁻). This makes them amphoteric – able to act as both acids and bases – which is why amino acids and proteins can act as buffers.
2. Peptide Bonds and Condensation |
Amino acids are linked by condensation reactions. The carboxyl group of one amino acid reacts with the amino group of another, a molecule of water is eliminated, and a peptide bond (a C–N bond) forms between the two residues. Two amino acids joined this way form a dipeptide.
H₂N–CH(R₁)–COOH + H₂N–CH(R₂)–COOH → H₂N–CH(R₁)–CO–NH–CH(R₂)–COOH + H₂O
The reaction is reversible. Hydrolysis of the peptide bond splits a polypeptide back into amino acids, and it is catalysed by proteases and peptidases during digestion. This is exactly why proteins must be digested before absorption: only free amino acids can cross the intestinal epithelium.
The peptide bond has partial double-bond character, which makes it planar and rigid. This restricts rotation and helps explain why polypeptide chains fold into such precise and reproducible shapes. The chain has direction: the free amino group end is the N-terminus and the free carboxyl end is the C-terminus.
Chains are named by length: dipeptide, tripeptide, oligopeptide (a few residues), and polypeptide (many residues). A functional protein may consist of a single polypeptide or of several polypeptides assembled together.
t a reactant, and make sure the C=O of the new peptide bond points towards the C-terminus.
3. Primary Structure |
The primary structure of a protein is the sequence (and number) of amino acids in its polypeptide chain(s), held together only by peptide bonds. The sequence is determined by the base sequence of a gene: DNA is transcribed into mRNA, which is translated at the ribosome.
Even a single substitution can change function dramatically. In sickle cell anaemia, glutamic acid at position 6 of the β-globin chain is replaced by valine. The replacement introduces a hydrophobic patch on the surface of haemoglobin, so the molecules stick together at low oxygen concentrations and form long fibres, distorting the red blood cell into a sickle shape.
The primary structure determines all higher levels of structure. All the information required for a polypeptide to fold into its final three-dimensional shape is contained in the sequence itself, although in cells molecular chaperones assist the folding process.
Exam tip: the mark scheme definition is precise – "the sequence of amino acids in a polypeptide chain". Answers such as "the shape of the protein" or "how the protein is arranged" score zero.
4. Secondary Structure: α-Helix and β-Pleated Sheet |
en bonds form between the partially negative oxygen of a C=O group and the partially positive hydrogen of an N–H group in the backbone – not between R groups.
In an α-helix, the chain coils into a spiral with hydrogen bonds running parallel to the axis of the helix. There are about 3.6 amino acid residues per complete turn, and the R groups project outwards from the helix, where they can interact with other molecules. Keratin is a classic α-helical fibrous protein.
In a β-pleated sheet, regions of the chain lie side by side in a zig-zag arrangement, with hydrogen bonds forming between adjacent strands. The strands may run in the same direction (parallel) or in opposite directions (antiparallel). R groups alternate above and below the plane of the sheet. Silk fibroin is rich in β-pleated sheets, and the interior of many globular proteins contains them.
Many proteins also contain random coil or loop regions that do not fit either pattern but are still essential for function, especially in forming active sites and binding sites.
Exam tip: if a question asks which bonds stabilise secondary structure, the answer is hydrogen bonds only. Mentioning disulfide bridges or ionic bonds here loses the mark.
5. Tertiary Structure and the Bonds Involved |
Tertiary structure is the further folding of the entire polypeptide chain into a precise, compact three-dimensional shape. It brings together amino acids that are far apart in the primary sequence, creating active sites, binding sites and recognition surfaces.
Tertiary structure is maintained by several types of bond and interaction:
- Hydrogen bonds between polar R groups | 极性 R 基团之间的氢键
- Ionic bonds between oppositely charged R groups, e.g. –NH₃⁺ and –COO⁻ | 带相反电荷 R 基团之间的离子键,如 –NH₃⁺ 与 –COO⁻
- Hydrophobic interactions and van der Waals forces between non-polar R groups | 非极性 R 基团之间的疏水相互作用与范德华力
- Disulfide bridges (–S–S–) between the R groups of two cysteine residues – covalent and the strongest of these links | 两个半胱氨酸残基 R 基团之间的二硫桥 (–S–S–),为共价键,是这些连接中最强的
- Peptide bonds, which hold the backbone together |
Individually, hydrogen bonds and ionic bonds are weak, but because there are so many of them, the overall shape is highly stable. In globular proteins dissolved in water, non-polar R groups tend to be buried in the interior to avoid contact with water, while polar and charged R groups face outwards – the hydrophobic core stabilises the fold.
Exam tip: examiners frequently ask for exactly three named interactions. Learn to write "hydrogen bonds between R groups, ionic bonds between charged R groups and disulfide bridges between cysteine residues", and add hydrophobic interactions for a high-level answer.
6. Quaternary Structure and Conjugated Proteins |
Quaternary structure is the way in which two or more polypeptide chains (subunits) associate to form a single functional protein. It may also involve non-protein prosthetic groups. Not every protein has a quaternary structure – myoglobin and lysozyme are single-chain proteins.
Haemoglobin is the classic example: four polypeptide chains (two α and two β) arranged around four haem groups, each containing an Fe²⁺ ion that can reversibly bind one oxygen molecule. Therefore one haemoglobin molecule can carry four oxygen molecules, and binding is cooperative – the first oxygen bound makes the next easier to bind, producing the characteristic S-shaped oxygen dissociation curve.
Proteins that contain a non-protein component are called conjugated proteins. Examples include haemoglobin (haem prosthetic group), glycoproteins (carbohydrate), lipoproteins (lipid), nucleoproteins (nucleic acid) and metalloproteins (metal ions).
Exam tip: quaternary structure always involves more than one polypeptide chain. A single chain with a haem group has tertiary structure plus a prosthetic group, but no quaternary structure.
7. Fibrous vs Globular Proteins |
Proteins can be broadly divided into fibrous and globular types. Fibrous proteins are
