Enzymes are one of the most frequently tested topics in GCSE Biology. This bilingual guide explains what enzymes are, why they are called biological catalysts, how the lock and key and induced fit models describe their action, and how temperature and pH change reaction rates. It also walks through the two classic practicals, catalase with hydrogen peroxide and the amylase starch-iodine test, then looks at everyday uses of enzymes and how to structure a full-mark exam answer.
What Are Enzymes: Catalysts and Active Sites
Enzymes are proteins, and occasionally RNA, produced by living cells. They speed up chemical reactions by lowering the activation energy, and they are not changed or used up by the reaction, which is why they count as catalysts. Each enzyme contains an active site, a specific pocket whose shape comes from the enzyme's folded three-dimensional structure. The substrate binds at the active site and is converted into products. Because the shape of the active site is unique, enzymes show high specificity: amylase in saliva only hydrolyses starch, never fats or proteins.
Lock and Key vs Induced Fit
The lock and key model describes the active site as a rigid shape, rather like a keyhole. Only a substrate whose shape is exactly complementary, the key, can slot in and bind, just as a key opens a lock. This model is simple and explains specificity well. The induced fit model is closer to what actually happens: as the substrate approaches, the enzyme changes shape slightly, and the active site wraps around the substrate like a glove, making catalysis more efficient. Exam questions often ask you to compare the two models and to state which one better explains evidence such as competitive inhibition, where an inhibitor with a similar shape to the substrate can also bind to the enzyme.
How Temperature Affects Enzyme Activity
As temperature rises, molecules move faster, so enzymes and substrates collide more often and the rate of reaction increases, roughly doubling for every 10°C rise. However, above the optimum temperature, about 37°C for human enzymes, extra heat energy breaks the hydrogen bonds that hold the enzyme's three-dimensional structure together. The enzyme denatures: its active site changes shape, the substrate can no longer bind, and the rate falls rapidly to zero. You may be asked to sketch the rate-temperature curve and explain the steep drop after the optimum. Remember that denaturation is permanent; cooling the mixture does not restore the enzyme.
四、pH 与最适条件 / pH and the Optimum Condition
Every enzyme has an optimum pH. Very high or very low hydrogen ion concentrations disturb the ionic and hydrogen bonds between the amino acids of the enzyme, so the enzyme denatures and stops working. Most human enzymes work best near neutral pH 7, but pepsin in the stomach has an optimum of about pH 2, while trypsin in the small intestine prefers around pH 8. A common exam question asks you to compare the two curves: the rate-temperature curve rises steadily and then collapses, whereas the rate-pH curve is bell-shaped and falls smoothly on both sides, because extreme pH in either direction denatures the enzyme.
Catalase and Hydrogen Peroxide
Hydrogen peroxide breaks down slowly into water and oxygen on its own. Catalase, found in large amounts in fresh potato and liver, speeds up this decomposition dramatically: 2H2O2 -> 2H2O + O2. In the practical, pieces of fresh potato are added to hydrogen peroxide solution, and a glowing splint is held at the mouth of the tube. The splint relights, showing that oxygen is being produced quickly. Boiled potato produces almost no bubbles because its enzymes have been denatured. To measure the rate, collect the gas in an upturned measuring cylinder and record the volume every 10 seconds; plotting a volume-time graph lets you compare rates using the slope. For a fair test, change only the factor you are investigating and keep temperature, pH, enzyme concentration, substrate concentration and enzyme volume constant.
The Amylase and Iodine Experiment
This investigation is the standard way to find the optimum temperature or pH for amylase. Starch turns blue-black when iodine solution is added, but the product maltose does not. Mix starch solution with amylase and, every 30 seconds, remove one drop of the mixture onto a white tile and add a drop of iodine. When the blue-black colour no longer appears, all the starch has been digested and you record the time taken. Repeating the experiment in water baths at 10, 20, 30, 40, 50 and 60°C shows that the fastest clearing time marks the optimum temperature. Always pre-warm the starch and the enzyme separately in the water bath before mixing them; otherwise you measure an average rate across several temperatures. One common trap is that the reaction continues inside each sample drop after it has been removed, so mix the drop with iodine immediately or cool it in an ice bath to stop the reaction.
Everyday Uses and an Exam-Style Answer
Enzymes are widely used at home and in industry. Biological washing powders contain proteases and lipases: proteases break down insoluble protein stains such as blood and egg into small soluble molecules, while lipases convert grease into glycerol and fatty acids, so the stains wash out more easily. These powders must never be used with boiling water, because the high temperature denatures the enzymes. In the food industry, pectinase is added to clarify fruit juice, and rennet, which contains enzymes, is used to make cheese. Here is a worked exam-style question: explain why amylase hardly digests starch at 60°C. A full-mark answer follows three steps. Firstly, 60°C is above the optimum temperature of this enzyme. Secondly, the high temperature breaks the hydrogen bonds holding the enzyme's three-dimensional structure, so the enzyme denatures. Thirdly, after denaturation the active site changes shape, starch can no longer bind to it, and the reaction almost stops. This step-by-step chain of reasoning matches every mark point on the mark scheme, and it is the standard structure for explanation questions in the biology paper.
Study Tips
1) Draw both models yourself and label the active site, the substrate and the products. 2) Keep two ideas separate: specificity comes from the shape of the active site, while reaction rate is controlled by temperature, pH and concentrations. 3) Learn the three tests well: a glowing splint relights with oxygen, iodine turns blue-black with starch, and biuret reagent detects protein; do not mix them up. 4) For graph questions, describe the trend first and then explain it, mentioning the optimum and the fact that denaturation is permanent. 5) In practical questions, state the control variables first because they are the easiest marks. 6) Memorise the bilingual pairs enzyme, active site, denature, optimum temperature, catalase and amylase so that gap-fill questions become free marks. 7) Before the exam, practise one complete enzyme practical question against the clock: write down the control variables, then the procedure and observations, then explain the result using the three-step logic chain above. Once this routine is automatic, you will not panic in the real paper.
