GCSE AQA Biology
## Cell Structure
All living organisms are made of cells, the basic building blocks of life. In GCSE AQA Biology, you need to know the difference between eukaryotic and prokaryotic cells. Eukaryotic cells — found in animals, plants, and fungi — contain a true nucleus where genetic material (DNA) is enclosed within a nuclear membrane. Prokaryotic cells, such as bacteria, lack a true nucleus; instead, their DNA floats freely in the cytoplasm as a single circular chromosome, sometimes accompanied by small rings called plasmids. This fundamental distinction is one of the most tested concepts in the entire GCSE Biology specification.
Animal cells contain a nucleus, cytoplasm, cell membrane, mitochondria, and ribosomes. Plant cells have all of these plus a rigid cell wall made of cellulose, a permanent vacuole filled with cell sap, and chloroplasts that carry out photosynthesis. Bacterial cells are much smaller and simpler: they have a cell wall (not made of cellulose), cell membrane, cytoplasm, and free-floating DNA. Some bacteria also have a slime capsule for protection and flagella for movement. When the exam asks you to compare cell types, always create a mental checklist of these organelles and their functions.
A common exam question asks you to calculate the magnification or actual size of a cell from a diagram. The formula is simple: Magnification equals Image size divided by Actual size. Always convert units consistently — remember that 1 millimetre equals 1000 micrometres, and 1 micrometre equals 1000 nanometres. Practice rearranging this formula and converting between millimetres, micrometres, and nanometres until it becomes second nature. These calculation questions are almost always worth 2 to 3 marks, so mastering them is a quick way to boost your overall grade.
## Cell Specialisation and Differentiation
As an organism develops, cells differentiate to become specialised for particular functions. Differentiation is the process by which a cell changes to become specialised. In animal cells, the ability to differentiate is lost at an early stage, but many plant cells retain this ability throughout their life. This is why plants can regrow from cuttings, while most animal tissues cannot regenerate in the same way.
Key specialised cells you must know for the exam include: sperm cells, which have a streamlined head, lots of mitochondria for energy, and an acrosome containing digestive enzymes to penetrate the egg; nerve cells, which are long and have branched endings to carry electrical impulses; muscle cells, which contain many mitochondria and special proteins that contract; root hair cells, which have a large surface area to absorb water and minerals; xylem cells, which are hollow tubes strengthened by lignin to transport water; and phloem cells, which have sieve plates to transport dissolved sugars around the plant. For each cell type, the examiner wants you to link the structural adaptation directly to its function — never just list features without explanation.
## Microscopy
Microscopes have been essential to our understanding of cell biology. The light microscope was developed in the 1590s and can magnify up to about 2000 times. It uses visible light and lenses to form an image. The electron microscope, developed in the 1930s, uses beams of electrons instead of light and can magnify up to two million times, with much higher resolution. Electron microscopes allow us to see subcellular structures like ribosomes, mitochondria cristae, and even individual molecules. While light microscopes let us observe living cells in real time, electron microscopes require dead specimens in a vacuum, which is an important limitation.
For the required practical, you must know how to prepare a microscope slide of onion epidermal cells or cheek cells. The steps are: place a drop of water or stain (such as iodine or methylene blue) on a clean slide, place the specimen in the liquid, lower a coverslip carefully at an angle to avoid air bubbles, and then observe under the microscope starting with the lowest power objective lens. Always draw your observations using a sharp pencil, label the visible structures, and include the magnification. Remember to start with the lowest magnification first to locate the specimen, then switch to higher magnifications for detailed observation.
## Cell Division
The cell cycle is the series of events that cells go through as they grow and divide. It consists of three main stages: interphase (where the cell grows, DNA replicates, and organelles are copied), mitosis (where the nucleus divides), and cytokinesis (where the cytoplasm and cell membrane divide to form two identical daughter cells). The cell cycle is carefully regulated, and uncontrolled cell division is what leads to cancer, making this topic directly relevant to understanding disease.
Mitosis is essential for growth, repair, and asexual reproduction. During mitosis, the chromosomes line up at the centre of the cell, and then spindle fibres pull them apart to opposite poles. Each new daughter cell receives an identical copy of the parent cell's DNA. In plants, mitosis occurs mainly in the meristems — the growing tips of roots and shoots. This is why plant growth is concentrated at the tips, and why cutting the tip of a root or shoot can affect a plant's growth pattern.
A common misconception is confusing mitosis with meiosis. Meiosis produces gametes (sperm and egg cells) with half the normal number of chromosomes, and is covered in the Inheritance topic of GCSE Biology, not Cell Biology. Make sure you do not mix these up in the exam, as examiners are particularly strict about this distinction.
## Transport in Cells
Substances move in and out of cells by three main processes: diffusion, osmosis, and active transport. Diffusion is the net movement of particles from an area of higher concentration to an area of lower concentration, down the concentration gradient. It is a passive process that requires no energy. Examples include oxygen diffusing from the alveoli into the blood, and carbon dioxide diffusing out of cells into the bloodstream. The larger the concentration gradient and the greater the surface area, the faster the rate of diffusion.
The rate of diffusion is affected by several factors: the concentration gradient (steeper gradient means faster diffusion), temperature (higher temperature provides more kinetic e
nergy, increasing diffusion rate), and surface area (a larger surface area allows more particles to cross at once). In the lungs, the alveoli have a large surface area, thin walls (one cell thick), and a rich blood supply — all adaptations that maximise the rate of gas exchange. The small intestine has similar adaptations with its villi and microvilli providing an enormous surface area for absorbing digested food molecules.
Osmosis is a special type of diffusion involving water molecules moving across a partially permeable membrane from a dilute solution to a more concentrated solution. In the required practical, you investigate osmosis by placing potato cylinders in different concentrations of sugar or salt solution, measuring the change in mass or length. If the solution is more dilute than the potato cells (hypotonic), water enters and the potato gains mass. If the solution is more concentrated (hypertonic), water leaves and the potato loses mass. In an isotonic solution, there is no net movement of water. Always calculate the percentage change rather than absolute change to allow fair comparison between samples.
Active transport, unlike diffusion and osmosis, requires energy from respiration in the form of ATP. It moves substances against the concentration gradient — from low to high concentration. Root hair cells use active transport to absorb mineral ions from the soil even when the concentration inside the cell is higher than in the surrounding soil. This is essential because mineral ions are often at very low concentrations in soil water, yet plants need them for healthy growth. The gut lining also uses active transport to absorb glucose when the concentration in the gut is lower than in the blood.
## Stem Cells
Stem cells are undifferentiated cells that can develop into different types of specialised cells. In humans, embryonic stem cells (from embryos) can differentiate into any type of cell — they are described as pluripotent. Adult stem cells (found in bone marrow and other tissues) can only differentiate into a limited range of cell types. In plants, meristem cells in the root and shoot tips are stem cells that can differentiate into any plant cell type throughout the plant's life, which is why plants can be cloned so easily.
Stem cells have important medical applications. They can be used to treat conditions such as diabetes (by producing insulin-secreting cells) and paralysis (by replacing damaged nerve cells). Bone marrow transplants, which contain adult stem cells, are already used to treat leukaemia. In plants, stem cells from meristems are used to clone rare plant species and produce large numbers of identical plants for commercial agriculture. Therapeutic cloning, where an embryo is created with the same genes as the patient to harvest stem cells, is an area of active research but also raises significant ethical questions.
There are ethical concerns surrounding the use of embryonic stem cells. Some people believe that human embryos have the potential for life and should not be destroyed for medical research. Others argue that the potential benefits of treating serious diseases outweigh these concerns. The exam may ask you to discuss both sides of this ethical debate, so be prepared to present balanced arguments. In the UK, stem cell research is strictly regulated by the Human Fertilisation and Embryology Authority, and only embryos up to 14 days old may be used for research purposes.
## Exam Tips and Common Mistakes
One of the most frequent errors in GCSE Cell Biology questions is confusing the cell wall and the cell membrane. Remember: the cell membrane controls what enters and leaves the cell in all cell types. The cell wall is an additional rigid layer found in plants, algae, and fungi (made of cellulose in plants and algae, chitin in fungi) that provides structural support. Bacteria also have a cell wall, but it is not made of cellulose. In exam answers, always be precise about which organisms have which structures.
Another common pitfall is stating that "mitochondria produce energy." This is incorrect — mitochondria are the site of aerobic respiration, which releases energy from glucose. Energy is never "produced" or "created"; it is released or transferred. Examiners are strict about this distinction. Use the phrase "mitochondria are the site of aerobic respiration, where energy is released" to earn full marks. Similarly, chloroplasts do not "make energy" — they absorb light energy to carry out photosynthesis, converting carbon dioxide and water into glucose and oxygen.
When describing osmosis, avoid saying "water moves to equalise concentrations." While the result is that concentrations equalise, the definition must state that water moves across a partially permeable membrane from a dilute to a more concentrated solution. Also, never use the word "turgid" to describe an animal cell — animal cells burst (lyse) in hypotonic solutions because they lack a cell wall. Only plant cells can become turgid and only plant cells undergo plasmolysis in hypertonic solutions.
Remember the three key steps for bacterial culturing practicals: Sterilise the inoculating loop and Petri dish using a Bunsen burner flame or autoclave to kill any unwanted microorganisms, Seal the dish with adhesive tape (but not fully — leave small gaps so oxygen can enter for aerobic respiration), and Store the dish upside down in an incubator at 25 degrees Celsius to prevent condensation from dripping onto the agar surface and disrupting the bacterial colonies. These steps are guaranteed marks if you recall them clearly.
For the osmosis required practical, always mention that you should blot the potato cylinders dry with a paper towel before measuring mass, to remove excess surface water that would affect the results. Also, calculate the percentage change in mass rather than just the absolute change, as this makes the results comparable across cylinders of different starting masses. When plotting the graph, the independent variable (concentration of solution) goes on the x-axis, and the percentage change in mass goes on the y-axis.
