GCSE Biology
Microscopy is one of the most fundamental skills in GCSE Biology. Whether you are sitting AQA, Edexcel, OCR or CIE, understanding how to use a microscope and prepare specimens is essential for both your practical exam and written paper.
## What is Microscopy? |
Microscopy is the technique of using microscopes to view objects and areas of objects that cannot be seen with the naked eye. The word "microscope" comes from the Greek words "mikros" meaning small and "skopein" meaning to look or see. In GCSE Biology, we primarily focus on the light microscope, which uses visible light and a system of lenses to magnify images of small samples.
The light microscope revolutionised biology when it was first developed in the late 16th century. For the first time, scientists could observe cells, microorganisms and the detailed structures of living things. Robert Hooke used an early microscope to observe cork cells in 1665, and this observation led to the development of cell theory. Today, microscopes remain a cornerstone of biological research, from diagnosing diseases to studying the intricate structures within cells.
## Parts of a Light Microscope |
Understanding each part of the microscope is the first step to using it correctly. Here are the key components you need to know for your GCSE exam.
The eyepiece lens is the lens at the top that you look through. It usually has a magnification of 10x, meaning it makes the image appear ten times larger than its actual size.
The objective lenses are the lenses closest to the specimen. A typical school microscope has three objective lenses: low power with 4x magnification, medium power with 10x magnification and high power with 40x magnification. The total magnification is calculated by multiplying the eyepiece magnification by the objective lens magnification. So if you are using a 10x eyepiece with a 40x objective lens, the total magnification is 400x.
The stage is the flat platform where you place your slides. The stage has clips to hold the slide in place, preventing it from moving during observation.
The coarse focus knob is the larger knob used to move the stage up and down to bring the specimen into general focus. You should only use the coarse focus knob with the low power objective lens. Using it with the high power lens can crack the slide and damage the lens.
The fine focus knob is the smaller knob used to fine-tune the focus and see the specimen in sharp detail. Use this with medium and high power objectives.
The diaphragm or iris controls the amount of light reaching the specimen. Adjusting the diaphragm can improve the contrast of the image. The light source or mirror provides illumination from below the stage.
## How to Use a Microscope |
The GCSE practical exam often asks you to describe the steps for using a microscope. Here is the standard procedure that examiners look for. GCSE
Step One: Place the microscope on a flat, stable surface near a power source if it has a built-in light. Plug in and switch on the light.
Step Two: Select the lowest power objective lens, which is usually 4x. Rotate the nosepiece until the 4x lens clicks into place. Always start with the lowest power because it gives the widest field of view and makes it easier to locate the specimen.
Step Three: Place your prepared slide on the stage and secure it with the stage clips. Make sure the coverslip is facing upwards and the specimen is centred over the hole in the stage where the light passes through.
Step Four: Use the coarse focus knob to raise the stage until the objective lens is close to the slide. Look from the side, not through the eyepiece, to avoid crashing the lens into the slide. When the lens is almost touching the slide, stop.
Step Five: Look through the eyepiece and slowly turn the coarse focus knob to lower the stage until the specimen comes into rough focus. Adjust the diaphragm to achieve the right brightness.
Step Six: Use the fine focus knob to bring the image into sharp, clear focus. You should now see the specimen clearly.
Step Seven: If you need higher magnification, carefully rotate the nosepiece to select the medium power 10x objective. Only use the fine focus knob from this point onward. Repeat for the high power 40x lens if needed, but be extremely careful as the working distance is very short.
Important safety note: Never use the coarse focus knob with the high power objective lens. The lens is so close to the slide that coarse adjustment can easily crack the coverslip and damage both the slide and the expensive objective lens.
## Preparing a Microscope Slide |
Slide preparation is a core practical skill tested in GCSE Biology. There are several methods depending on the type of specimen you are examining.
For a wet mount, which is the most common preparation method, you need a glass slide, a coverslip, the specimen, water and a pipette. Place a drop of water in the centre of a clean glass slide using a pipette. Place your specimen gently into the water droplet. To add the coverslip, hold it at a 45-degree angle against the slide, with one edge touching the water droplet. Slowly lower the coverslip over the specimen. This angled technique prevents air bubbles from being trapped under the coverslip, which would obstruct your view of the specimen.
For staining, many biological specimens are transparent and colourless, making them difficult to see under the microscope. Applying a stain adds contrast by colouring specific parts of the cell. Common stains in GCSE Biology include iodine solution for staining starch in plant cells, which turns blue-black in the presence of starch. Methylene blue is used for staining animal cells, particularly the nucleus. To stain a specimen, place a drop of stain at one edge of the coverslip, then draw a piece of filter paper across the opposite edge. The stain will be pulled under the coverslip, replacing the water and staining the cells.
For preparing a thin section, specimens like plant stems or leaves need to be cut into very thin slices to allow light to pass through. Use a razor blade or scalpel to cut th
e thinnest possible cross-section. Place the section in a drop of water on the slide and cover with a coverslip. The thinner the section, the better you can see individual cells and their structures.
## Magnification & Resolution |
Students often confuse magnification and resolution, but these are two very different concepts that GCSE examiners frequently test.
Magnification is how much larger an image appears compared to the actual size of the object. The formula you must memorise is: Magnification equals Image Size divided by Actual Size. If an image of a cell measures 50 millimetres and the actual cell measures 0.05 millimetres, the magnification is 50 divided by 0.05, which equals 1000 times. You must be comfortable rearranging this formula to calculate any of the three variables.
Resolution, on the other hand, is the ability to distinguish between two separate points that are very close together. Think of it as the sharpness or clarity of the image. A high resolution means you can see two adjacent points as distinct and separate. A low resolution means they blur together into one fuzzy blob. The resolution of a light microscope is limited by the wavelength of visible light. The maximum resolution of a light microscope is approximately 200 nanometres, which means two points closer than 200 nanometres apart cannot be seen as separate.
The key difference is that you can increase magnification indefinitely by using stronger lenses, but beyond a certain point, the image just becomes bigger and blurrier without revealing any new detail. This is called empty magnification. Resolution determines the useful limit of magnification. A light microscope can magnify up to about 1500 times usefully, beyond which empty magnification occurs.
## Electron Microscopes |
The development of electron microscopes in the 1930s overcame the resolution limitations of light microscopes by using beams of electrons instead of visible light. Since electrons have a much shorter wavelength than visible light, electron microscopes can achieve much higher resolution, down to about 0.1 nanometres, which allows scientists to see the internal structures of organelles like mitochondria and chloroplasts in extraordinary detail.
There are two main types of electron microscopes you need to know for GCSE. The Transmission Electron Microscope or TEM produces two-dimensional images by passing electrons through a very thin specimen. TEM can achieve the highest magnification and resolution, revealing the internal ultrastructure of cells. The Scanning Electron Microscope or SEM produces three-dimensional images of the specimen surface by scanning it with a beam of electrons. SEM images show the surface topography in striking detail.
However, electron microscopes have important limitations. They are extremely expensive, require a vacuum environment which means specimens cannot be living, involve complex specimen preparation, and produce black and white images that are later false-coloured. Light microscopes remain essential because they can observe living specimens in real time, are portable and affordable. Both types of microscope remain important tools in modern biology.
## Common Exam Questions |
GCSE examiners frequently test microscopy through specific question types. Understanding these patterns will help you score highly. GCSE
Calculation questions are extremely common. You may be given an image of a cell with a scale bar and asked to calculate the actual size or the magnification. Always remember to convert units correctly: 1 millimetre equals 1000 micrometres, and 1 micrometre equals 1000 nanometres. Write the formula first, substitute the numbers, and show your working clearly for method marks even if you make a calculation error.
Method questions ask you to describe how to prepare a slide or use a microscope to observe a specimen. Examiners want logical sequential steps with key details. Mention holding the coverslip at 45 degrees to avoid air bubbles, starting on low power, and using only fine focus with high power. Include safety precautions.
Comparison questions ask for the differences between light and electron microscopes. Create a balanced response covering magnification, resolution, specimen requirements, cost, portability and image type. Use comparative language.
Diagram questions may ask you to label parts of a microscope or draw what you observe through the eyepiece. Practise drawing clear biological diagrams with a sharp pencil, using continuous lines, adding labels with straight ruled lines, and including a title and magnification.
