遗传学在IB生物中的重要性 / Why Genetics Matters in IB Biology
Genetics is one of the largest and most heavily examined topics in IB Biology Core. It spans the entirety of Topic 3 (Genetics), covering gene and chromosome structure, meiosis, Mendelian inheritance, molecular genetics, and genetic modification techniques. For Standard Level students, genetics accounts for roughly 15 teaching hours. Higher Level students delve deeper into the detailed stages of meiosis as well as advanced concepts such as gene linkage and recombination. Mastering genetics is not just a prerequisite for exam success -- it is the gateway to understanding modern biology, from precision medicine to CRISPR gene editing.
基因、DNA与染色体 / Genes, DNA and Chromosomes
A gene is a segment of DNA located at a specific position (locus) on a chromosome that codes for a protein or functional RNA, thereby controlling an organism's traits. The DNA double helix consists of two antiparallel polynucleotide strands, with base pairing following the A-T, G-C rule. In eukaryotes, DNA associates with histone proteins and compacts into chromosomes. Human somatic cells contain 46 chromosomes (23 pairs), of which 22 pairs are autosomes and one pair are sex chromosomes. In IB exams, students must distinguish between three easily confused terms: gene (the basic unit of heredity), allele (an alternative form of a gene), and genome (the complete set of genetic information in an organism). Contrasting homologous chromosomes with sister chromatids is another common pitfall: homologous chromosomes originate from each parent and carry the same loci but potentially different alleles, whereas sister chromatids are two identical copies connected by a centromere after DNA replication -- a distinction repeatedly tested in meiosis questions.
Mendelian Genetics
Mendel discovered the Law of Segregation and the Law of Independent Assortment through his pea plant hybridization experiments, laying the foundation of modern genetics. The Law of Segregation states that each individual carries two alleles, and these alleles segregate during gamete formation so that each gamete contains only one allele. The Law of Independent Assortment states that alleles at different loci assort independently during gamete formation. Common IB exam tasks include drawing genetic diagrams for monohybrid and dihybrid crosses, and using Punnett squares to calculate genotypic and phenotypic ratios. It is important to note that dominance is not absolute: dominant alleles typically encode functional proteins, while recessive alleles often encode non-functional proteins or are not expressed at all. Codominance and incomplete dominance extend the Mendelian model and frequently appear in IB HL exams, often tied to real-world cases such as blood group inheritance.
分子遗传学:从DNA到蛋白质 / Molecular Genetics: From DNA to Protein
The central dogma of molecular genetics describes the flow of genetic information: DNA replicates semi-conservatively, is transcribed into mRNA, and is translated into protein. DNA replication occurs during S phase, with key enzymes including helicase and DNA polymerase. DNA polymerase synthesizes new strands only in the 5' to 3' direction, resulting in continuous synthesis of the leading strand and discontinuous synthesis of the lagging strand via Okazaki fragments. During transcription, RNA polymerase recognizes the promoter region and synthesizes mRNA using the template strand. Introns are excised during RNA splicing, and exons are joined to form mature mRNA. Translation occurs on ribosomes, where tRNA anticodons base-pair with mRNA codons, and the ribosome catalyzes peptide bond formation to produce a polypeptide chain. IB exams require students to explain how gene mutations affect protein structure and function, with sickle cell anemia -- caused by a single base substitution -- serving as a classic example of the cascading effects of a point mutation. In terms of exam weight, genetics accounts for roughly 15 percent of Paper 1 questions; Paper 2 data-based questions frequently feature electrophoresis gels or pedigree charts as prompts, and the Section B long-answer question almost always includes an 8-mark genetics item each year. Students opting for Paper 3 Option B (Biotechnology) should bridge knowledge across both modules, flexibly applying the principles of PCR and gel electrophoresis to justify the experimental design of genetic analysis investigations.
Inheritance Patterns: Beyond Mendel
The Mendelian model cannot account for all inheritance phenomena. In sex-linked inheritance, genes located on the X chromosome exhibit distinctive transmission patterns. Since males possess only one X chromosome, recessive X-linked alleles are always expressed in males, whereas females require homozygosity for the recessive allele to show the trait. Red-green colour blindness and haemophilia are classic examples of human X-linked recessive disorders. Multiple allele systems, such as the ABO blood group, demonstrate codominance -- where IA and IB alleles are both expressed -- in population genetics. In polygenic inheritance, multiple genes collectively determine a continuous trait, such as human skin colour or height, and these traits follow a normal distribution in populations. Linked genes reside on the same chromosome and do not assort independently; instead, they are transmitted together in parental combinations. However, crossing over during meiosis can produce recombinant gametes, and IB HL exams frequently require students to infer recombination frequency between genes from offspring ratios.
Biotechnology and Genetic Modification
The IB Biology syllabus covers several key biotechnologies. The Polymerase Chain Reaction (PCR) uses Taq DNA polymerase to rapidly amplify specific DNA fragments in vitro through three cyclic steps -- denaturation, annealing, and extension -- and is widely applied in forensics and disease diagnosis. Gel electrophoresis separates DNA mixtures by fragment size; DNA migrates toward the positive electrode due to its negatively charged phosphate groups, with smaller fragments moving faster, enabling DNA fingerprinting and paternity testing. In genetic modification (GMO) technology, restriction endonucleases cut DNA at specific recognition sites to produce sticky ends, DNA ligase joins the target gene with a plasmid vector, and the recombinant plasmid is introduced into host bacteria via transformation. Genetically modified organisms have important applications in agriculture (Bt insect-resistant corn) and medicine (insulin production). IB exams frequently require students to analyse the ethical implications of GMOs, including environmental impact, food safety, and gene patenting controversies.
IB考试策略与常见误区 / IB Exam Strategy and Common Pitfalls
State that DNA replication is semi-conservative"只需说明结论,而"Explain semi-conservative replication"则需要描述Meselson-Stahl实验的证据。
In genetics exams, the command term determines the depth of response required. Distinguish between "State" (simply present a fact) and "Explain" (require causal mechanisms): for example, "State that DNA replication is semi-conservative" needs only the conclusion, while "Explain semi-conservative replication" requires describing the evidence from Meselson and Stahl's experiment. When drawing genetic diagrams, always label parental genotypes, gamete types, and offspring ratios. HL students must additionally annotate crossing over events and recombination frequencies. Common calculation errors include confusing genotypic ratios with phenotypic ratios, and overlooking the differential probabilities between male and female offspring in sex-linked inheritance. We recommend creating concept comparison tables during revision -- juxtaposing meiosis vs mitosis, transcription vs translation, and chromosomal mutations vs gene mutations side by side helps systematically organize the knowledge structure. When tackling human pedigree chart questions, first determine the inheritance pattern (autosomal dominant/recessive, X-linked dominant/recessive), then analyse individual genotypes one by one.
