AQA A-Level Biology Topic 3: Organisms Exchange Substances with Their Environment is one of the most content-dense units in the entire specification. Worth approximately 20% of Paper 1 marks, this topic spans gas exchange across five organism groups, digestive system biochemistry, haemoglobin oxygen transport, the cardiac cycle, and mass flow in plants. The key to mastering Exchange is understanding that every adaptation you encounter, from lamellae to villi to sieve plates, follows one unifying principle: surface area maximised, diffusion distance minimised, concentration gradient maintained.
1. Surface Area to Volume Ratio
As organisms increase in size, their surface area to volume ratio (SA:V) decreases dramatically. A cube with 1 cm sides has SA:V = 6:1; a cube with 10 cm sides drops to 0.6:1. This fundamental geometric constraint is why large multicellular organisms cannot rely on simple diffusion across their outer surface: the diffusion pathway becomes too long and the relative surface area too small to supply every cell with sufficient oxygen and remove carbon dioxide efficiently.
Key application in exams: AQA frequently asks you to calculate SA:V ratios and explain the consequences for heat exchange in larger organisms (e.g., why elephants have large ears, why shrews need to eat constantly). Always link your answer to the three principles: SA:V ratio, diffusion distance, and metabolic rate. For gas exchange systems specifically, remember that each specialised surface (alveoli, gill lamellae, tracheoles) is a solution to the SA:V problem: folded, branched, or flattened structures that create a large exchange surface within a compact body.
2. Gas Exchange Across Five Organism Groups
AQA requires you to compare gas exchange adaptations in single-celled organisms, insects, fish, plants, and humans. Each system is a variation on the same theme: maximise diffusion rate using Fick's Law.
Insects
Tracheal system: spiracles open into tracheae which branch into tracheoles ending directly at muscle cells. Air moves via rhythmic abdominal pumping; tracheole fluid draws in water by osmosis during high activity, increasing gas dissolution surface. Spiracles close to reduce water loss, a key terrestrial adaptation.
Fish
Gill filaments with lamellae at right angles. Countercurrent flow: blood flows through lamellae in the opposite direction to water flow over gills. This maintains a concentration gradient along the entire exchange surface, achieving up to 80% oxygen extraction. Buccal-opercular pumping drives continuous unidirectional ventilation.
Plants 植物
Dicotyledonous leaves: CO2 enters through stomata in the lower epidermis, diffuses through air spaces in spongy mesophyll to reach palisade cells. Stomatal opening controlled by guard cell turgor (K+ ion influx lowers water potential, water enters, guard cells curve open). Large internal surface area provided by intercellular air spaces.
Humans 人类
Trachea→bronchi→bronchioles→alveoli. Inspired air: external intercostals contract, ribs move up/out, diaphragm contracts and flattens, thoracic volume increases, pressure drops below atmospheric. Expiration is largely passive (elastic recoil). Alveolar epithelium is one cell thick, surrounded by extensive capillary network creating massive SA:V.
Exam precision matters: When describing insect tracheal systems, specify that tracheoles are not lined with chitin (unlike tracheae), allowing direct gas exchange with muscle cells. For fish, never just say "countercurrent flow" without explaining that blood and water flow in opposite directions, which maintains a steady diffusion gradient along the full length of the lamella. For plants, note that stomata are mainly on the lower epidermis of dicot leaves, reducing water loss while permitting CO2 entry. For humans, be precise about the two sets of intercostal muscles: external intercostals contract during inspiration; internal intercostals contract during forced expiration.
3. Digestion and Absorption
Digestion breaks large insoluble polymers into small soluble monomers that can cross epithelial membranes. AQA splits this into mechanical digestion (teeth, stomach churning) and chemical digestion (hydrolysis reactions catalysed by specific enzymes), each with a precise syllabus location.
Carbohydrates 碳水
Salivary amylase (mouth): starch→maltose (pH 6.5-7.0). Pancreatic amylase (duodenum): continues starch hydrolysis (pH 7.5-8.0). Membrane-bound disaccharidases on ileum epithelial microvilli: maltase, sucrase, lactase hydrolyse disaccharides→monosaccharides. Monosaccharides absorbed via co-transport with Na+ (sodium-glucose linked transporter SGLT1).
Proteins
Endopeptidases (pepsin in stomach pH 2, trypsin in duodenum pH 7.5): hydrolyse internal peptide bonds → shorter polypeptides. Exopeptidases: hydrolyse terminal peptide bonds → dipeptides and single amino acids. Dipeptidases on ileum epithelial membrane: hydrolyse dipeptides→amino acids. Amino acids absorbed via Na+ co-transport.
Lipids 脂类
Bile salts (produced in liver, stored in gall bladder): emulsify large lipid droplets→smaller droplets by reducing surface tension, increasing surface area for lipase action. Pancreatic lipase: hydrolyses triglycerides→monoglycerides + 2 fatty acids. Micelles: monoglycerides + fatty acids + bile salts form water-soluble micelles that carry lipids to ileum epithelial surface where they diffuse in. Inside epithelial cells, triglycerides reform, then packaged into chylomicrons → lacteals → lymphatic system.

(3) Rich capillary network in each villus maintains a steep concentration gradient by rapidly transporting absorbed products away. (4) Lacteals in villus core absorb chylomicrons from lipid digestion. (5) Membrane-bound enzymes (disaccharidases, dipeptidases) complete digestion at the point of absorption. (6) Mitochondria in epithelial cells provide ATP for active Na+ transport, driving the co-transport of glucose and amino acids.

4. Mass Transport in Animals
Once substances cross exchange surfaces, they must be distributed throughout the body. AQA focuses on three interconnected systems: haemoglobin oxygen loading/unloading, the closed double circulatory system, and tissue fluid formation.
Haemoglobin
Quaternary structure: 4 polypeptide chains (2 alpha, 2 beta), each with a haem group containing Fe2+ that binds O2. Cooperative binding: binding of first O2 changes conformation, making subsequent binding easier, producing the sigmoidal dissociation curve. The Bohr effect: increased CO2 lowers pH (via carbonic acid), shifting the curve right (lower O2 affinity), unloading more O2 in respiring tissues. Foetal haemoglobin has a higher O2 affinity than adult haemoglobin (curve shifted left), allowing O2 transfer across the placenta.
Cardiac Cycle
Atrial systole: atria contract, AV valves open, blood forced into ventricles. Ventricular systole: ventricles contract from apex upwards, AV valves snap shut (first heart sound "lub"), semilunar valves open, blood ejected into aorta and pulmonary artery. Diastole: both chambers relax, semilunar valves close (second sound "dub"), passive filling begins. Key vessels: aorta (left ventricle→body), pulmonary artery (right ventricle→lungs), vena cava (body→right atrium), pulmonary vein (lungs→left atrium).
Tissue Fluid 组织液
At the arteriole end of capillaries: hydrostatic pressure (from ventricular contraction) exceeds oncotic pressure (from plasma proteins), forcing fluid out through capillary pores → tissue fluid formed. At the venule end: hydrostatic pressure drops (resistance in capillary bed), oncotic pressure now exceeds hydrostatic, drawing water back in (90% reabsorbed). Remaining 10% drains into the lymphatic system and returns to blood via subclavian veins.

5. Mass Transport in Plants
Plants have two transport systems: xylem for water and dissolved mineral ions (unidirectional, roots→leaves) and phloem for organic solutes like sucrose (bidirectional, source→sink). AQA requires you to explain the mechanisms driving both, with precise terminology for top marks.
Cohesion-Tension Theory
Water evaporates from mesophyll cells into leaf air spaces, diffuses out through stomata (transpiration). This creates a negative pressure (tension) at the top of the xylem column. Due to cohesion (hydrogen bonds between water molecules) and adhesion (water binding to xylem lignin walls), the entire water column is pulled up as a continuous unbroken stream. Root pressure provides a minor contribution at night. Factors increasing transpiration rate: higher temperature (more kinetic energy), lower humidity (steeper water potential gradient), higher wind speed (removes boundary layer of humid air), and higher light intensity (stomata open wider for photosynthesis).
Mass Flow Hypothesis
At the source (e.g., leaf mesophyll): sucrose is actively loaded into companion cells (H+ gradient established by proton pumps, sucrose enters via co-transporter proteins), then diffuses through plasmodesmata into sieve tube elements. This lowers the water potential in sieve tubes, causing water to enter from adjacent xylem by osmosis. The resulting hydrostatic pressure drives bulk flow toward the sink (e.g., root tips, developing fruits), where sucrose is actively unloaded and used or stored. Water then returns to xylem via osmosis, completing the pressure gradient.
Evidence for mass flow: (1) Aphid stylets inserted into sieve tubes exude sap under pressure, confirming hydrostatic flow. (2) Radioactive 14C labelling of CO2 shows movement of labelled sucrose from source leaves to sink tissues. (3) Metabolic poisons that inhibit ATP production stop translocation, confirming active loading and unloading are energy-dependent.
