1. Cell Biology
Cells are the basic structural and functional units of all living organisms. There are two fundamentally different cell types you must distinguish clearly in exams. Eukaryotic cells contain a true nucleus where the genetic material (DNA) is enclosed within a nuclear membrane. Examples include all animal cells, plant cells, and fungal cells. Prokaryotic cells lack a nucleus -- instead, their DNA floats freely in the cytoplasm as a single circular chromosome, sometimes accompanied by small rings of DNA called plasmids. Bacteria are the primary example.
Animal cell organelles: Nucleus (contains genetic material and controls cell activities), cytoplasm (site of most chemical reactions), cell membrane (selectively permeable barrier controlling what enters and leaves), mitochondria (site of aerobic respiration, releasing energy from glucose), ribosomes (site of protein synthesis).
Plant cells have all of the above plus: Cell wall (made of cellulose, provides structural support), chloroplasts (contain chlorophyll, site of photosynthesis), permanent vacuole (filled with cell sap, maintains turgor pressure).
Cell specialisation: Sperm cells (acrosome with digestive enzymes, haploid nucleus, mitochondria for swimming, tail/flagellum). Nerve cells (long axon, myelin sheath, dendrites for connections). Muscle cells (contain many mitochondria, can contract due to protein fibres). Root hair cells (large surface area, no chloroplasts, thin cell wall). Xylem cells (hollow tubes, lignin-reinforced walls, dead at maturity). Phloem cells (sieve plates, companion cells).
2. Microscopy and Required Practical
The light microscope uses visible light and lenses to magnify specimens up to roughly 2000 times. The electron microscope, invented in the 1930s, uses beams of electrons and can magnify up to 2,000,000 times with far higher resolution, revealing subcellular structures like ribosomes and internal mitochondrial membranes.
Magnification formula: Magnification = Image size / Actual size. Remember unit conversions: 1cm = 10mm, 1mm = 1000 micrometres, 1 micrometre = 1000 nanometres. Always present your answer in the unit requested by the question. Standard form is frequently used: a bacterium measuring 2 micrometres is 2 x 10 to the power of negative 6 metres.
Required Practical: Prepare an onion epidermis slide. Peel a thin layer of epidermal tissue from inside an onion layer, place it flat on a drop of water on a glass slide, add a drop of iodine solution (stains the nucleus and cell wall), lower a coverslip carefully at an angle to avoid air bubbles, then observe under low and high power objectives. Draw what you see using a sharp pencil -- label with straight ruled lines, include magnification, and never shade or colour.
3. Cell Division and Stem Cells
Mitosis is the process of cell division that produces two genetically identical daughter cells. It occurs during growth, repair, and asexual reproduction. The stages: Interphase (DNA replicates, cell grows), Prophase (chromosomes condense and become visible, nuclear membrane breaks down), Metaphase (chromosomes line up at the equator), Anaphase (spindle fibres pull chromatids to opposite poles), Telophase (nuclear membranes reform, cytokinesis completes the split).
Stem cells are undifferentiated cells that can divide to produce more stem cells or differentiate into specialised cells. Embryonic stem cells are pluripotent -- they can become almost any cell type. Adult stem cells (e.g. in bone marrow) are multipotent -- limited to certain cell types. In plants, meristems in root and shoot tips contain stem cells that differentiate throughout the plant's life. Therapeutic cloning can produce patient-matched embryonic stem cells, avoiding rejection. Ethical debates centre on the destruction of embryos in research.
4. Enzymes 酶
Enzymes are biological catalysts -- globular proteins that speed up metabolic reactions without being used up or changed permanently. Each enzyme has a specific active site that is complementary in shape to its specific substrate. This is the lock and key model: the substrate fits into the active site like a key into a lock, forming an enzyme-substrate complex. The reaction occurs and products are released. More recent thinking uses the induced fit model, where the active site slightly changes shape to accommodate the substrate.
Temperature: As temperature rises, enzyme and substrate molecules gain kinetic energy and collide more frequently. The rate increases up to an optimum (around 37 degrees Celsius for human enzymes). Beyond this, bonds in the enzyme break, the active site changes shape irreversibly -- the enzyme is denatured and the rate drops sharply.
pH: Each enzyme has an optimum pH. Pepsin (stomach) works best at pH 2. Amylase (saliva) works best at pH 7. If pH moves too far from the optimum, hydrogen and ionic bonds break, denaturing the enzyme.
Required Practical: Investigate the effect of pH on amylase. Mix amylase solution with starch solution and a buffer (pH 5, 6, 7, or 8). Every 30 seconds, transfer a drop to a spotting tile well with iodine solution. Iodine turns blue-black if starch is present, orange-brown if absent. Record the time taken for starch to disappear completely at each pH. Plot a graph of pH against rate (1/time).
5. Transport, Osmosis, and Organisation
Diffusion is the net movement of particles from a region of higher concentration to a region of lower concentration, down the concentration gradient. It is a passive process requiring no energy. Rate is increased by: larger concentration gradient, higher temperature (more kinetic energy), and larger surface area to volume ratio. In the lungs, oxygen diffuses from alveoli into blood capillaries; carbon dioxide diffuses in the opposite direction. In leaves, carbon dioxide diffuses through stomata into the mesophyll.
Osmosis is the diffusion of water molecules across a partially permeable membrane from a dilute solution (high water potential) to a more concentrated solution (low water potential). If an animal cell is placed in pure water, it will swell and burst (lysis) because it has no cell wall. Plant cells in pure water become turgid (swollen but supported by the cell wall) -- this is ideal. In concentrated sugar solution, plant cells become flaccid and eventually plasmolyzed (cell membrane pulls away from the cell wall).
Required Practical: Osmosis in potato cylinders. Cut equal-sized potato cylinders, blot dry, weigh, then place in different sucrose concentrations (0M to 1M). After 30 minutes, remove, blot, and reweigh. Calculate percentage change in mass = (final mass - initial mass) / initial mass x 100. Negative change means water left the potato (hypertonic solution). Positive change means water entered (hypotonic solution). Plot a graph and find where the line crosses zero -- this is the concentration inside the potato cells.
6. Bioenergetics: Photosynthesis
The balanced symbol equation: 6CO2 + 6H2O --> C6H12O6 + 6O2 (in the presence of light and chlorophyll). Photosynthesis is an endothermic reaction -- it takes in energy from sunlight, transferred by chloroplasts containing chlorophyll.
Plants are photoautotrophs: they make their own food using light. The glucose produced is used immediately for respiration, converted to starch for storage (insoluble, so it does not affect water potential), used to make cellulose for cell walls, combined with nitrate ions to produce amino acids for protein synthesis, and turned into lipids (fats and oils) for seed storage.
Limiting factors: Light intensity, carbon dioxide concentration, and temperature. Only one factor limits the rate at any given time. On a graph, the rate increases as the limiting factor increases, then plateaus when something else becomes limiting. Greenhouses are designed to optimise all factors: artificial lighting (extend daylight), paraffin heaters (release CO2 and heat), and ventilation (prevent overheating). The economics are important -- the cost of providing extra light, heat, or CO2 must be less than the extra revenue from increased yield.
Required Practical: Effect of light intensity on photosynthesis. Place a piece of pondweed (Elodea or Cabomba) in a boiling tube with water and sodium hydrogen carbonate (releases CO2). Count the number of oxygen bubbles produced per minute at different distances from a light source. Light intensity is proportional to 1 divided by distance squared -- an inverse square relationship. Alternatively, measure the volume of gas collected in a gas syringe over time.
7. Respiration and Metabolism
Aerobic respiration is exothermic and occurs continuously in the mitochondria: C6H12O6 + 6O2 --> 6CO2 + 6H2O + energy (ATP). All organisms respire aerobically whenever oxygen is available. The energy released is used for: muscle contraction, maintaining body temperature (in mammals and birds), active transport, building larger molecules (anabolism), and cell division.
Anaerobic respiration in animals: Glucose --> Lactic acid + energy. This occurs during vigorous exercise when the oxygen supply to muscles is insufficient. The buildup of lactic acid causes muscle fatigue and cramps. Oxygen debt is the extra oxygen needed after exercise to oxidise accumulated lactic acid into carbon dioxide and water (in the liver). This is why you continue breathing heavily after exercise stops.
Anaerobic respiration in plants and yeast: Glucose --> Ethanol + Carbon dioxide + energy. This is fermentation, exploited in bread-making (CO2 makes dough rise, ethanol evaporates during baking) and brewing (ethanol is the alcohol in beer and wine). Industrial fermentation uses yeast in anaerobic conditions with a carbohydrate substrate.
Metabolism is the sum of all chemical reactions in a cell or organism. It includes: conversion of glucose to starch, glycogen, and cellulose; formation of lipid molecules from a glycerol molecule and three fatty acid molecules; protein synthesis from amino acids; respiration; and photosynthesis. The liver is the metabolic hub, performing detoxification, protein synthesis, and glycogen storage (insulin triggers conversion of glucose to glycogen; glucagon triggers the reverse).
8. Genetics and Inheritance
DNA is a polymer made up of two strands forming a double helix. Each strand consists of repeating nucleotide units: a phosphate group, a deoxyribose sugar, and one of four bases -- Adenine (A), Thymine (T), Cytosine (C), and Guanine (G). Complementary base pairing: A pairs with T (two hydrogen bonds), C pairs with G (three hydrogen bonds). A gene is a section of DNA that codes for a sequence of amino acids, which makes a specific protein.
Protein synthesis: Transcription takes place in the nucleus -- the DNA double helix unzips and one strand serves as a template to produce a complementary mRNA molecule. The mRNA carries the code out of the nucleus to the cytoplasm. Translation occurs at ribosomes -- the mRNA code is read in triplets (codons), each codon matching a specific amino acid delivered by tRNA. The amino acids join to form a polypeptide chain that folds into a functional protein.
Genetics vocabulary: Allele (different version of the same gene), genotype (the combination of alleles), phenotype (the observable characteristic), homozygous (two identical alleles for a trait, e.g. BB or bb), heterozygous (two different alleles, e.g. Bb), dominant (allele expressed when present even in one copy), recessive (allele expressed only when two copies are present).
Monohybrid inheritance: Use Punnett squares. Example: Cystic fibrosis is caused by a recessive allele f. Cross heterozygous parents Ff x Ff: offspring genotypes FF (25%, normal), Ff (50%, carrier), ff (25%, has cystic fibrosis). Ratios: 3:1 phenotype (3 normal, 1 affected), 1:2:1 genotype. Polydactyly (extra digits) is caused by a dominant allele D -- only one parent needs the allele for offspring to be affected.
Family pedigree charts: Squares represent males, circles represent females. Shaded shapes show affected individuals. You may be asked to work out genotypes from a pedigree and calculate probabilities.
9. Evolution by Natural Selection
Charles Darwin proposed the theory of evolution by natural selection after his voyage on HMS Beagle, particularly influenced by observations of finches on the Galapagos Islands. The process: Within any species, there is genetic variation caused by mutations. Individuals with characteristics better suited to the environment are more likely to survive and reproduce (survival of the fittest). These advantageous alleles are passed to the next generation. Over many generations, the frequency of these alleles increases, and the species evolves.
Antibiotic resistance is a classic example of natural selection observable within a human lifetime. In a bacterial population, a random mutation may give one bacterium resistance to an antibiotic. When antibiotics are used, non-resistant bacteria die, but the resistant bacterium survives and reproduces. Over time, the population of resistant bacteria increases. MRSA (methicillin-resistant Staphylococcus aureus) is a dangerous example. This is why doctors prescribe antibiotics only when necessary and insist patients complete the full course.
Fossils provide evidence for evolution: they show organisms that no longer exist, document gradual changes over geological time, and reveal transitional forms. However, the fossil record is incomplete because: soft-bodied organisms rarely fossilise, geological activity can destroy fossils, and many fossils remain undiscovered. Additional evidence comes from: comparative anatomy (pentadactyl limb in vertebrates), antibiotic resistance in bacteria, and DNA analysis showing genetic relationships.
