A-Level AQA Biology 细胞生物学全攻略 / Mastering Cells for AQA A-Level Biology

Cells is a core module in AQA A-Level Biology Paper 1, covering eukaryotic cell structure, prokaryotic cells, viruses, membrane transport, mitosis, and microscopy. These concepts are not only heavily examined but also form the foundation for later topics including biochemistry, immunology, and genetics. This guide systematically covers 5 core knowledge points to help you secure top marks in the exam.

Eukaryotic Cell Structure

The AQA specification requires you to know the structure and function of every organelle in eukaryotic cells. The nucleus is the largest organelle, enclosed by a double nuclear envelope with nuclear pores that allow mRNA and ribosomal subunits to pass through. Inside, the nucleolus is the site of rRNA synthesis and ribosome assembly. Mitochondria are the sites of cellular respiration, with a double membrane structure — the outer membrane is smooth while the inner membrane folds inward to form cristae, dramatically increasing the surface area for ATP synthesis. Chloroplasts, found only in plant cells and algae, are the sites of photosynthesis and contain stacked thylakoids forming grana. Rough ER has ribosomes attached to its surface and is involved in protein folding and transport; Smooth ER is responsible for lipid synthesis. The Golgi apparatus receives vesicles from the ER, modifying, sorting, and packaging proteins. Lysosomes contain hydrolytic enzymes for intracellular digestion and autophagy. Ribosomes, composed of rRNA and protein, are where translation occurs — 80S ribosomes exist in eukaryotic cytoplasm while 70S ribosomes are found in chloroplasts and mitochondria, providing key evidence for the endosymbiotic theory of eukaryotic cell origin.

Prokaryotic Cells and Viruses

The key differences between eukaryotic and prokaryotic cells are a high-frequency exam topic in AQA. Prokaryotic cells are much smaller than eukaryotic cells and lack membrane-bound organelles. They contain circular DNA free in the cytoplasm and small DNA rings called plasmids. Prokaryotes have 70S ribosomes (smaller than the 80S ribosomes in eukaryotes) and a cell wall made of peptidoglycan (also called murein). Some prokaryotes have a capsule for protection against the immune system, a flagellum for movement, and pili for conjugation to transfer plasmids. Viruses are acellular infectious particles consisting of a nucleic acid core (DNA or RNA) enclosed in a protein capsid. Some viruses such as HIV also have a lipid envelope derived from the host cell membrane. Viruses lack ribosomes, lack cytoplasm, and cannot carry out independent metabolic activity, which is why they are not considered living organisms. They must invade host cells and use the host's enzymes and ribosomes to replicate. HIV is a retrovirus that contains reverse transcriptase, an enzyme that reverse-transcribes its RNA genome into DNA and integrates it into the host chromosome.

Cell Membrane Structure and Transport

The cell membrane follows the fluid mosaic model, consisting of a phospholipid bilayer. Phospholipid molecules have a hydrophilic phosphate head and hydrophobic fatty acid tails, spontaneously arranging into a bilayer in water. Protein molecules are embedded within the phospholipid bilayer — intrinsic proteins span the entire membrane while extrinsic proteins are attached only to the membrane surface. Cholesterol molecules sit between phospholipids, regulating membrane fluidity: at high temperatures they restrict phospholipid movement to enhance stability, while at low temperatures they prevent tight packing to maintain fluidity. Glycoproteins and glycolipids play roles in cell recognition and cell signalling.

There are four main mechanisms of transport across cell membranes. Simple diffusion is the passive movement of small, non-polar molecules (such as O2 and CO2) from high to low concentration through the phospholipid bilayer. Facilitated diffusion uses channel proteins or carrier proteins to help polar molecules and ions move down their concentration gradient without requiring ATP. Osmosis is the diffusion of water molecules through a partially permeable membrane from a region of high water potential to a region of low water potential. Active transport uses carrier proteins to move molecules against their concentration gradient, requiring energy from ATP hydrolysis — for example, the sodium-potassium pump. Co-transport is especially important in AQA — take glucose absorption as an example: sodium ions are actively transported out of epithelial cells by the sodium-potassium pump, establishing a sodium concentration gradient; sodium ions then diffuse back down this gradient through a co-transporter protein, simultaneously bringing glucose molecules into the cell.

Mitosis and the Cell Cycle

The cell cycle is divided into interphase and the mitotic phase. Interphase accounts for about 90% of the cell cycle and is divided into three stages: G1 (Gap 1) where the cell grows and synthesises proteins and organelles; S (Synthesis) where DNA replication occurs and each chromosome becomes two sister chromatids joined at the centromere; and G2 (Gap 2) where the cell continues to grow and checks that DNA replication is correct. The mitotic phase includes mitosis and cytokinesis. Mitosis itself has four stages: prophase where chromosomes condense and become visible, the nuclear envelope breaks down, and spindle fibres begin to form; metaphase where chromosomes align at the equator (metaphase plate) and spindle fibres attach to centromeres; anaphase where centromeres divide and spindle fibres shorten, pulling sister chromatids to opposite poles; and telophase where chromosomes decondense, nuclear envelopes reform, and the spindle breaks down. During cytokinesis, animal cells form a cleavage furrow to divide into two, while plant cells form a cell plate at the equator that develops into a new cell wall. Mitosis produces two genetically identical daughter cells and is used for growth, repair, and asexual reproduction.

The AQA exam may ask you to estimate the duration of cell cycle stages — this can be done by observing the proportion of cells in each stage, since the longer a cell spends in a stage, the higher the probability of observing it in that stage. For example, if in a root tip sample 80 out of 200 cells are observed in mitotic stages, and mitosis is known to last approximately 2 hours, the full cell cycle can be estimated at about 5 hours.

Microscopy Techniques

Microscopy is the core of AQA Required Practical 2. Light microscopes use visible light and a lens system to magnify specimens, with a maximum resolution of about 200 nm. Magnification = eyepiece magnification x objective lens magnification. Resolution refers to the ability to distinguish between two adjacent points and is limited by the wavelength of light. Actual size = image size / magnification. Transmission electron microscopes (TEM) use an electron beam passing through ultra-thin stained specimens, achieving resolution down to 0.5 nm and revealing internal organelle structure — but specimens must be in a vacuum and are dead. Scanning electron microscopes (SEM) scan the specimen surface and detect reflected electrons, producing three-dimensional images with a resolution of about 3-10 nm.

Cell fractionation is the experimental technique for separating organelles. Tissue is ground in a cold isotonic buffer — cold to reduce enzyme activity, isotonic to prevent osmotic damage, and buffered to maintain stable pH. The homogenate is centrifuged at increasing speeds: low speed (1000g) pellets nuclei and unbroken cells, medium speed (10000g) pellets mitochondria and chloroplasts, and high speed (100000g) pellets ribosomes and membrane fragments. The supernatant from each centrifugation is used for the next round, and the pellet contains the target organelles.
