📚 Enzymes: Structure, Function and Factors Affecting Activity |
Enzymes are biological catalysts that speed up chemical reactions in living organisms without being used up themselves. They are essential for life, controlling everything from digestion to DNA replication. This revision guide covers the key concepts required for Edexcel IGCSE Biology, including enzyme structure, the active site, and the factors that affect enzyme activity.
1. Chemical Nature of Enzymes |
Enzymes are proteins, made up of long chains of amino acids folded into specific three-dimensional shapes. This folding is critical because the shape determines the enzyme's function. The unique sequence and arrangement of amino acids create a special region called the active site.
2. The Active Site |
The active site is the part of an enzyme where the substrate molecule binds. It is usually a small pocket or groove on the surface of the enzyme. The shape of the active site is complementary to the shape of the substrate, so only specific substrates can fit perfectly. This is why enzymes are highly specific, acting on only one or a few substrates.
3. Lock and Key vs Induced Fit Model |
To explain enzyme specificity, the lock and key model suggests that the active site is rigid like a lock, and only the correctly shaped substrate (key) can fit. In contrast, the induced fit model proposes that the active site is flexible and changes shape slightly to mould around the substrate after binding. Both models highlight the importance of shape matching.
4. Enzyme-Substrate Complex |
When a substrate binds to the active site, a temporary enzyme-substrate complex forms. During this time, the enzyme lowers the activation energy of the reaction, allowing the substrate to be converted into product. After the reaction, the product is released, and the enzyme is unchanged and ready to catalyse another reaction.
5. Factors Affecting Enzyme Activity: Temperature |
Temperature affects the kinetic energy of molecules. As temperature rises, molecules move faster, and enzyme-substrate collisions become more frequent, so the rate of reaction increases. However, above the optimum temperature, the vibrations break the bonds holding the enzyme together, causing the active site to change shape. This process is called denaturation, and the enzyme can no longer function.
Optimum temperature for most human enzymes ≈ 37°C
Each enzyme has an optimum pH at which it works best. At extreme pH values, the charges on amino acids are altered, which can disrupt ionic and hydrogen bonds. This changes the shape of the active site and causes denaturation. For example, pepsin in the stomach works best at pH 2, while most other human enzymes work best at pH 7.
7. Factors Affecting Enzyme Activity: Substrate Concentration |
When substrate concentration increases, the rate of an enzyme-catalysed reaction rises because more substrate molecules are available to collide with active sites. However, this effect is limited: once all active sites are occupied, the enzyme is saturated and the rate reaches a plateau. Only adding more enzyme can increase the rate further.
8. Enzyme Concentration |
Increasing enzyme concentration also increases the rate of reaction, provided substrate is in excess. More enzyme molecules mean more active sites are available to bind substrate. As with substrate concentration, the rate will eventually plateau if substrate becomes limiting.
9. Inhibitors |
Inhibitors are substances that reduce the activity of an enzyme. Competitive inhibitors structurally resemble the substrate and compete for the active site; their effect can be overcome by increasing substrate concentration. Non-competitive inhibitors bind to another part of the enzyme, changing the shape of the active site. Their effect cannot be overcome simply by adding more substrate.
10. Enzymes in Living Organisms |
Enzymes are involved in every metabolic reaction. Examples include:
- Amylase – breaks down starch into maltose in the mouth and small intestine.
- Catalase – decomposes hydrogen peroxide into water and oxygen in cells.
- Protease – hydrolyses proteins into amino acids in the stomach and small intestine.
- Lipase – breaks down fats into fatty acids and glycerol in the small intestine.
11. Practical Applications: Immobilised Enzymes |
Immobilised enzymes are attached to an inert support, making them easier to reuse. They are widely used in industry, for example in making fructose syrup from glucose using glucose isomerase. Immobilisation allows continuous processing, saves time and reduces costs, and the enzyme remains stable at higher temperatures.
12. Exam Tips |
In Edexcel IGCSE Biology exams, always refer to 'active site', 'complementary shape' and 'denaturation' when explaining enzyme specificity or the effect of temperature and pH. When interpreting graphs, identify the optimum point and describe the plateau. Use the correct terms: 'substrate', 'product', 'enzyme-substrate complex', and state that enzymes are proteins.
