Organic synthesis is the planned construction of target molecules from simpler starting materials using a sequence of chemical reactions. In Edexcel A-Level Chemistry, this topic brings together reactions of alkenes, halogenoalkanes, alcohols, carbonyl compounds, carboxylic acids, esters, amines, nitriles and aromatic compounds. You must be able to design synthetic routes, choose appropriate reagents and conditions, and justify each step using yield and atom economy.
1. What Is Organic Synthesis? |
Organic synthesis is not just memorising single reactions. It is about linking functional group transformations into a logical route. An Edexcel exam question may give a starting material and a target molecule, then ask you to identify missing intermediates, reagents and conditions.
ic acid → ester. You should practise writing equations for each forward and backward step.
2. Core Synthetic Routes |
A large number of Edexcel organic synthesis questions are built around the alkene–halogenoalkane–alcohol–carbonyl–carboxylic acid–ester pathway. Each step requires specific reagents and conditions, so precise wording matters.
| Halogenoalkane + NaOH(aq) | 卤代烷 + NaOH 水溶液 | Heat under reflux | 加热回流 | Alcohol | 醇 |
| Primary alcohol + acidified K₂Cr₂O₇ | 伯醇 + 酸性 K₂Cr₂O₇ | Distil immediately | 立即蒸馏 | Aldehyde | 醛 |
| Primary alcohol + excess acidified K₂Cr₂O₇ | 伯醇 + 过量酸性 K₂Cr₂O₇ | Heat under reflux | 加热回流 | Carboxylic acid | 羧酸 |
| Carboxylic acid + alcohol | 羧酸 + 醇 | Concentrated H₂SO₄, reflux | 浓 H₂SO₄,回流 | Ester | 酯 |
CH₂=CH₂ + HBr → CH₃CH₂Br
CH₃CH₂Br + NaOH(aq) → CH₃CH₂OH + NaBr
CH₃CH₂OH + [O] → CH₃CHO + H₂O
CH₃CHO + [O] → CH₃COOH
CH₃COOH + CH₃CH₂OH ⇌ CH₃COOCH₂CH₃ + H₂O
3. Functional Group Interconversions |
A functional group interconversion chart is the most useful revision tool for organic synthesis. You must know which functional groups can be converted directly into others, and which conversions require first going through a halogenoalkane or alcohol.
For example, an alkene can be converted directly into an alcohol by steam hydration with concentrated H₃PO₄, or into a halogenoalkane by addition of HBr. However, an alkane cannot be converted directly into an alcohol; it must first undergo free-radical substitution to form a halogenoalkane.
4. Oxidation and Reduction Sequences |
Oxidation and reduction are central to building up carbonyl and alcohol functional groups. Primary alcohols can be partially oxidized to aldehydes if the product is distilled out immediately, because aldehydes have lower boiling points than carboxylic acids.
If primary alcohols are heated under reflux with excess acidified K₂Cr₂O₇, oxidation continues to the carboxylic acid. Secondary alcohols give ketones, and tertiary alcohols resist oxidation because they lack a hydrogen atom on the carbon bearing the –OH group.
CH₃CH₂OH + [O] → CH₃CHO + H₂O
CH₃CHO + [O] → CH₃COOH
Reduction is equally important. Sodium borohydride (NaBH₄) is a milder reducing agent used to reduce aldehydes and ketones to alcohols. Lithium aluminium hydride (LiAlH₄) is more powerful and can reduce carboxylic acids, esters and nitriles.
5. Carboxylic Acids and Derivatives |
Carboxylic acids are versatile intermediates because they can form esters, acyl chlorides and amides. Acyl chlorides are especially useful in synthesis: they react readily with alcohols to make esters and with ammonia or amines to make amides under mild conditions.
Using an acyl chloride instead of a carboxylic acid often gives a higher yield for esterification because the reaction is not reversible and does not require strong acid heating. However, acyl chlorides are moisture-sensitive and must be handled carefully.
CH₃COCl + CH₃CH₂OH → CH₃COOCH₂CH₃ + HCl
Esters can be hydrolysed back to the parent acid and alcohol under acidic conditions, or to the carboxylate salt under alkaline conditions. Alkaline hydrolysis is irreversible and often gives a better yield of the alcohol.
6. Nitrogen-Containing Compounds |
Nitriles are key intermediates for extending a carbon chain by one carbon atom. A halogenoalkane reacts with potassium cyanide
