Cambridge International A Level Biology is a content-heavy yet highly rewarding subject. The Coursebook by Mary Jones et al. provides comprehensive coverage, but many students struggle to translate textbook knowledge into exam success. This article pinpoints the most frequently assessed topics, breaks down tricky concepts, and highlights the command words examiners love to use. You will learn how to structure answers, apply knowledge to unfamiliar contexts, and avoid common pitfalls across the entire syllabus.
1. Cell Structure and Function |
Electron micrographs and diagrams of eukaryotic cells are staple exam material. You must be able to identify and annotate organelles such as the nucleus, rough and smooth endoplasmic reticulum (ER), Golgi apparatus, mitochondria, chloroplasts, and lysosomes. Be precise about the functions: for example, the rough ER synthesises and transports proteins due to its attached ribosomes, while the smooth ER is involved in lipid synthesis and detoxification. Examiners often ask you to compare prokaryotic and eukaryotic cells, so memorise key differences: prokaryotes lack a membrane-bound nucleus and organelles, have 70S ribosomes, and contain a circular DNA molecule in the nucleoid region. Plant cells additionally possess a large permanent vacuole, a cellulose cell wall, and chloroplasts.
Beyond structure, understand the principles of cell theory and the use of magnification calculations. The formula M = I/A size) is frequently tested. Always convert all measurements to the same unit, typically micrometres (µ he requested format. When drawing cells from a micrograph, use a sharp pencil, draw clear outlines, and never shade – the exam requires a line diagram with correct proportions.
2. Biological Molecules |
The four major classes of biological molecules – carbohydrates, lipids, proteins, and nucleic acids – are foundational. Carbohydrates: be able to distinguish monosaccharides (glucose, fructose, galactose), disaccharides (maltose, sucrose, lactose), and polysaccharides (starch, glycogen, cellulose). Learn the glycosidic bond formation through condensation reactions and breakage by hydrolysis. Examiners frequently ask about the relationship between structure and function: for instance, starch’s coiled helix makes it compact and ideal for energy storage in plants, while cellulose’s straight chains with hydrogen bonds between adjacent molecules give the cell wall high tensile strength.
y acids is a classic mark. An unsaturated fatty acid contains at least one C=C double bond, causing a kink in the hydrocarbon chain and thus making the lipid more fluid at room temperature. Phospholipids, with their hydrophilic phosphate heads and hydrophobic fatty acid tails, are the basis of cell membranes. Proteins: understand the four structural levels – primary (amino acid sequence), secondary (α‑helix and β‑ tiple polypeptide chains). The biuret test for proteins and the emulsion test for lipids are simple but easy marks if described in full detail.
3. Enzymes | 酶
Enzymes are globular proteins that act as biological catalysts. Their specificity arises from the precise shape of the active site. The induced‑fit model is now favoured over the lock‑and‑key hypothesis: the active site is not entirely complementary to the substrate initially, but moulds around it upon binding, stressing bonds and lowering activation energy. Be ready to explain how the initial rate of an enzyme‑controlled reaction is measured and how substrate concentration, enzyme concentration, temperature, and pH affect this rate. Always relate changes to kinetic energy, frequency of effective collisions, and denaturation of tertiary structure.
A common exam question presents a graph and asks you to identify and justify the limiting factor at different points on the curve. For example, at low substrate concentration, the rate increases linearly with concentration because there are many free active sites; at saturation, all active sites are occupied, so adding more substrate has no effect. Examiners also love to include a competitive vs non‑competitive inhibitor comparison. Competitive inhibitors bind to the active site and can be overcome by increasing substrate concentration, while non‑competitive inhibitors bind at an allosteric site, changing the shape of the active site permanently, so Vmax is reduced and cannot be restored by adding more substrate.
4. Cell Membranes and Transport |
The fluid mosaic model describes the membrane as a phospholipid bilayer with embedded proteins, glycoproteins, and cholesterol (in animal cells). The phospholipids are free to move laterally, giving the membrane fluidity, while cholesterol regulates fluidity by reducing phospholipid movement at high temperatures and preventing tight packing at low temperatures. You must distinguish between diffusion (passive, down a concentration gradient), facilitated diffusion (passive, via channel or carrier proteins), osmosis (net movement of water through a partially permeable membrane from a region of higher water potential to lower water potential), and active transport (against a concentration gradient, requiring ATP and carrier proteins).
ₛ + ψₚ, where ψₛ is solute potential (always negative or zero) and ψₚ is pressure potential (positive or zero). In plant cells, turgid cells have a pressure potential of roughly +500 kPa, while plasmolysed cells have a pressure potential of 0. Questions often present data from experiments with potato strips or red blood cells in different solutions: be able to describe the expected observations and explain them in terms of water potential and membrane permeability.
5. The Mitotic Cell Cycle |
The cell cycle comprises interphase (G₁, S, G₂) and mitotic phase (mitosis and cytokinesis). DNA replicates during S phase, producing two identical chromatids held together by a centromere. Mitosis is divided into prophase, metaphase, anaphase, and telophase, and is essential for growth, repair, and asexual reproduction. Examiners want you to describe the behaviour of chromosomes in each stage, not just name them. For example, during metaphase, chromosomes line up on the equator of the spindle, with spindle fibres attached to centromeres. During anaphase, centromeres split and sister chromatids are pulled to opposite poles by the shortening of spindle fibres.
Cancer is a common context question. You should link it to uncontrolled cell division caused by mutations in proto‑oncogenes and tumour suppressor genes. Be able to explain the roles of checkpoints in the cell cycle and how the loss of checkpoint control allows cells with damaged DNA to divide.
6. Nucleic Acids and Protein Synthesis |
ach containing a pentose sugar, a phosphate group, and a nitrogenous base. DNA is a double‑stranded antiparallel helix with complementary base pairing (A=T, C≡G) held by hydrogen bonds; RNA is usually single‑stranded and contains uracil instead of thymine. DNA replication is semi‑conservative, as proven by the Meselson– lymerase unwinds the DNA, builds a complementary mRNA strand using free RNA nucleotides; the mRNA then leaves through nuclear pores. Translation occurs on ribosomes in the cytoplasm: tRNA molecules, with specific anticodons, bring amino acids to the mRNA; peptide bonds form between adjacent amino acids to produce a polypeptide chain.
Gene mutations – substitution, insertion, deletion – can alter the amino acid sequence and thus protein structure. Sickle cell anaemia is a must‑know example: a single base substitution in the gene for the β‑globin chain of haemoglobin changes the codon from GAG to GTG, resulting in glutamic acid being replaced by valine. This causes haemoglobin molecules to aggregate, distorting red cells into a sickle shape, which reduces oxygen‑carrying efficiency and blocks capillaries.
7. Transport in Plants |
Xylem transports water and dissolved mineral ions from roots to shoots. The cohesion‑tension theory explains the upward movement: transpiration at the leaves generates a negative pressure (tension) that pulls water up the xylem as a continuous column, held together by cohesion between water molecules and adhesion to xylem walls. You must be able to describe the pathway of water from the soil through root hair cells, across the cortex (apoplast and symplast pathways), through the endodermis (where the Casparian strip blocks the apoplast, forcing water into the symplast), and into the xylem. Phloem translocates sucrose and amino acids from sources to sinks. The pressure‑flow (mass flow) hypothesis states that active loading of sucrose at the source reduces water potential, causing water to enter by osmosis, increasing hydrostatic pressure, which pushes phloem contents to areas of lower pressure at the sink, where sucrose is unloaded.
Exam questions often provide data from potometers or ringing experiments, requiring you to evaluate evidence for the transport models. Always link your explanations to water potential gradients and pressure differences.
8. Transport in Mammals |
The mammalian circulatory system is a closed, double circulation. The heart is myogenic: the sinoatrial node (SAN) initiates the heartbeat, the atrioventricular node (AVN) delays the impulse to allow atrial systole to finish before ventricular systole, and the Purkyne fibres carry the impulse to the ventricle apex for coordinated contraction. You should be able to interpret ECG traces showing the P wave (atrial depolarisation), QRS complex (ventricular depolarisation), and T wave (ventricular repolarisation). Blood pressure changes throughout the circulatory system are common graph‑based questions: explain why pressure is high in the aorta and arteries but drops sharply in the arterioles and capillaries due to increased resistance and leakage of fluid.
Haemoglobin and oxygen dissociation curves are essential. The sigmoidal shape results from cooperative binding: binding of the first O₂ molecule increases haemoglobin’s affinity for subsequent O₂ molecules. The Bohr effect describes how high CO₂ concentration lowers pH, shifting the curve to the right and causing haemoglobin to release more oxygen in respiring tissues. Fetal haemoglobin has a higher oxygen affinity than adult haemoglobin, shifting its curve to the left, enabling efficient oxygen transfer across the placenta.
9. Gas Exchange and Smoking |
Mammalian gas exchange takes place in the alveoli. The walls are one cell thick, lined with a film of water and surfactant (reducing surface tension and preventing collapse), and are surrounded by a dense network of capillaries. The steep concentration gradients for O₂ and CO₂ are maintained by ventilation and blood flow. In humans, breathing involves the diaphragm and intercostal muscles: during inspiration, the external intercostals contract and the diaphragm flattens, increasing thoracic volume and decreasing pressure, drawing air in. Be prepared to label a spirometer trace and calculate tidal volume, vital capacity, breathing rate, and minute ventilation.
The effects of smoking on the respiratory system are a key application. Tar destroys cilia and goblet cells, leading to mucus accumulation and smoker’s cough; it also contains carcinogens that cause lung cancer. Nicotine constricts arterioles and increases platelet stickiness, raising the risk of cardiovascular disease. Carbon monoxide binds irreversibly to haemoglobin, reducing the oxygen‑carrying capacity of the blood. These should be linked to chronic obstructive pulmonary disease (COPD), including chronic bronchitis and emphysema, where alveolar walls break down, reducing surface area for gas exchange.
10. Infectious Diseases and Immunity |
Pathogens include bacteria, viruses, fungi, and protoctists. Cholera, malaria, tuberculosis, and HIV/AIDS are the specific diseases required by the syllabus. For each, know the causative organism, mode of transmission, and a relevant detail: for example, Vibrio cholerae produces a toxin that causes excessive secretion of chloride ions into the small intestine, drawing water by osmosis and causing severe diarrhoea. Plasmodium (malaria) has a complex life cycle involving both a mosquito vector and human liver and blood stages. The immune response can be non‑specific (inflammation, phagocytosis by neutrophils and macrophages) or specific (B and T lymphocytes). B cells produce antibodies, leading to humoral immunity, while T cells provide cell‑mediated immunity. Be able to describe the clonal selection theory: a specific antigen selects a pre‑existing lymphocyte, which clones into many effector and memory cells.
Vaccination and antibiotic resistance are heavily examined. Explain how vaccines stimulate the primary immune response and produce memory cells so that upon secondary exposure, pathogen is eliminated before symptoms develop and how herd immunity works. For antibiotic resistance, be clear that mutations produce resistant alleles, and the overuse of antibiotics creates a selection pressure that increases the frequency of these alleles in bacterial populations.
11. Energy and Respiration |
Respiration is the process by which organic molecules are oxidised to release energy in the form of ATP. The four stages are glycolysis, the link reaction, the Krebs cycle, and oxidative phosphorylation. Glycolysis occurs in the cytoplasm and yields a net gain of 2 ATP and 2 reduced NAD (NADH) per glucose. The link reaction and Krebs cycle take place in the mitochondrial matrix: pyruvate is decarboxylated and dehydrogenated, producing CO₂ and reduced coenzymes (NADH and FADH₂). The vast majority of ATP is produced by oxidative phosphorylation along the inner mitochondrial membrane, where the electron transport chain generates a proton gradient that drives chemiosmosis via ATP synthase.
0.9. Be ready to calculate RQ from given volumes and interpret the results. Anaerobic respiration in mammals produces lactate; in yeast, ethanol and CO₂ are produced. The importance of regenerating NAD⁺ to allow glycolysis to continue is a classic hinge point for explanation questions.
12. Photosynthesis |
Photosynthesis occurs in chloroplasts and consists of the light‑dependent reactions (in the thylakoid membranes) and the light‑independent Calvin cycle (in the stroma). In the light‑dependent stage, chlorophyll absorbs light energy, which is used to split water (photolysis), producing O₂, protons, and electrons. The electrons pass through an electron transport chain, generating ATP (by chemiosmosis) and reducing NADP to NADPH. The Calvin cycle uses ATP and NADPH to fix CO₂ into carbohydrate. The key enzyme is Rubisco, which catalyses the carboxylation of ribulose bisphosphate (RuBP) to form two molecules of glycerate‑3‑phosphate (GP), which are then reduced to triose phosphate (TP). Some TP is used to regenerate RuBP, while the rest is converted to glucose, starch, or other organic molecules.
Limiting factors of photosynthesis – light intensity, carbon dioxide concentration, and temperature – are investigated with aquatic plants like Elodea. When explaining experimental traces, always refer to the Calvin cycle and the balance between GP and TP concentrations. For example, suddenly removing CO₂ causes RuBP to accumulate and GP to fall because the carboxylation step is blocked.
