📚 Edexcel Biology 6.1 Cellular Control | Edexcel
Cellular control refers to the complex set of mechanisms that allow a cell to switch genes on or off, adjust levels of protein synthesis, and coordinate activities such as the cell cycle and apoptosis. In Edexcel A Level Biology, this topic focuses on how gene expression is regulated at transcriptional, post-transcriptional, translational and post-translational levels, and how failure of these controls can lead to diseases such as cancer. A strong grasp of cellular control is essential for understanding cell differentiation, responses to environmental signals, and the molecular basis of inheritance.
1. Overview of Cellular Control |
Every somatic cell in a multicellular organism contains the same DNA, yet cells become specialised because only a subset of genes is expressed in each cell type. Cellular control determines which genes are transcribed into mRNA and which proteins are ultimately produced. This selective gene expression is the basis of differentiation, homeostasis and development. Control can occur at many stages of the central dogma, from chromatin remodelling to protein degradation.
Gene expression can be controlled at the transcriptional level by transcription factors that bind to DNA, at the post-transcriptional level by RNA processing and degradation, at the translational level by initiation factors, and at the post-translational level by protein modification. Understanding these control points helps explain how identical genotypes can produce many different phenotypes.
2. Gene Structure and Transcription Initiation |
In eukaryotes, a typical protein-coding gene consists of exons and introns. Upstream of the coding region lies the promoter, a DNA sequence where RNA polymerase and general transcription factors assemble to initiate transcription. Enhancer sequences can be located farther away and increase the rate of transcription, while silencer sequences decrease it. The interaction of these regulatory elements with specific transcription factors determines whether a gene is switched on or off in a given cell.
Transcription initiation begins when general transcription factors bind to the TATA box within the promoter, forming a transcription initiation complex. Activator proteins bound to enhancers then loop the DNA to contact the mediator complex, which stabilises RNA polymerase II at the promoter. Repressor proteins can block activator binding or recruit histone deacetylases, making the chromatin less accessible.
3. Transcription Factors and Promoters |
Transcription factors are proteins that bind to specific DNA sequences to control the rate of transcription. They can be activators, which increase transcription, or repressors, which decrease it. Each transcription factor contains a DNA-binding domain that recognises a particular motif in the promoter or enhancer, and an activation or repression domain that interacts with other proteins. In multicellular organisms, combinations of transcription factors generate precise patterns of gene expression.
For example, the transcription factor p53 is activated by DNA damage and promotes the expression of genes involved in cell cycle arrest and DNA repair. If p53 is mutated, damaged cells may continue to divide, increasing the risk of cancer. This illustrates how a single transcription factor can coordinate a whole cellular response pathway.
4. The lac Operon in Prokaryotes |
In prokaryotes, gene expression is often organised into operons. The lac operon in E. coli is a classic example of transcriptional control. It contains three structural genes: lacZ, lacY and lacA, which code for β-galactosidase, lactose permease and galactoside transacetylase respectively. These enzymes are needed to import lactose and break it down into glucose and galactose.
The lac operon is controlled by the lac repressor and the catabolite activator protein (CAP). When lactose is absent, the lac repressor binds to the operator and blocks RNA polymerase from transcribing the structural genes. When lactose is present, it is converted to allolactose, which binds to the lac repressor and changes its shape so it can no longer bind the operator. This allows transcription to proceed. Glucose levels also regulate the operon: low glucose leads to high cAMP, which binds CAP and stimulates transcription.
No lactose → repressor bound → transcription OFF | 无乳糖 → 阻遏蛋白结合 → 转录关闭
After transcription, eukaryotic pre-mRNA must be processed before it can be translated. A 5' cap and a poly-A tail are added for stability and export, and introns are removed by a spliceosome. Alternative splicing allows a single gene to produce multiple mRNA variants by joining exons in different combinations. This greatly increases the diversity of the proteome without increasing the number of genes.
For instance, the human gene DSCAM can generate thousands of different mRNA isoforms through alternative splicing. Errors in splicing can lead to genetic disorders such as spinal muscular atrophy. The cell can also regulate splicing by producing splicing enhancer or silencer proteins that promote or inhibit the use of particular splice sites.
6. Epigenetic Modifications |
Epigenetics involves heritable changes in gene expression that do not involve changes to the DNA sequence itself. The two main epigenetic mechanisms are DNA methylation and histone modification. DNA methylation usually occurs at CpG dinucleotides, where a methyl group is added to cytosine. This often represses transcription by preventing transcription factors from binding or by recruiting repressor complexes.
Histone modification includes acetylation, methylation and phosphorylation of histone tails. Histone acetylation, catalysed by histone acetyltransferases (HATs), neutralises the positive charge on lysine residues and loosens the interaction between histones and DNA, making the chromatin more accessible for transcription. Histone deacetylases (HDACs) remove acetyl groups and promote a more condensed, repressed state.
7. RNA Interference and Gene Silencing | RNA
RNA interference (RNAi) is a post-transcriptional mechanism that uses small RNA molecules to silence gene expression. Small interfering RNAs (siRNAs) and microRNAs (miRNAs) are produced from double-stranded RNA or hairpin precursors by the enzyme Dicer. One strand of the small RNA is incorporated into the RNA-induced silencing complex (RISC), which then pairs with complementary mRNA sequences.
If the base pairing is fully complementary, RISC cleaves the mRNA, leading to its degradation. If the pairing is only partially complementary, translation is repressed. RNA interference is used by cells to regulate gene expression and also acts as a defence mechanism against viruses. In the laboratory, synthetic siRNAs can be used to knock down specific genes to study their function.
8. Translational and Post-translational Control |
Gene expression can also be regulated at the level of translation. The availability of initiation factors, the length of the poly-A tail, and the presence of regulatory proteins binding to the 5' or 3' untranslated regions all influence how efficiently an mRNA is translated. Some mRNAs are stored in cytoplasmic granules and only translated when the cell receives a specific signal.
Once a polypeptide is synthesised, post-translational modifications can activate, deactivate, stabilise or destabilise it. Common modifications include phosphorylation, glycosylation, ubiquitination and proteolytic cleavage. For example, many enzymes are synthesised as inactive precursors called zymogens and only become active after cleavage. Ubiquitination tags proteins for destruction by the proteasome, providing rapid control of protein levels.
9. Cell Cycle Checkpoints and Apoptosis |
The cell cycle is tightly controlled by cyclins, cyclin-dependent kinases (CDKs) and checkpoint proteins. The G₁ checkpoint checks for DNA damage and cell size; the G₂ checkpoint ensures all DNA has been replicated correctly; the spindle checkpoint in metaphase ensures all chromosomes are attached to the spindle. If conditions are not met, the cell cycle is arrested until repairs are made, or the cell may undergo apoptosis.
Apoptosis is programmed cell death, a controlled process that removes damaged, infected or unnecessary cells without causing inflammation. It is triggered by internal signals such as DNA damage or by external signals such as cytokines. Caspase enzymes are activated in a cascade, leading to cell shrinkage, chromatin condensation and formation of membrane-bound apoptotic bodies that are cleared by phagocytes. Failure of apoptosis can contribute to cancer, while excessive apoptosis is seen in neurodegenerative diseases.
10. Mutations and Cancer: Loss of Control |
Cancer is fundamentally a disease of uncontrolled cell division caused by mutations in genes that regulate the cell cycle and apoptosis. Two main classes of cancer genes are oncogenes and tumour suppressor genes. Oncogenes are mutated forms of proto-oncogenes that normally promote cell division in a controlled way; when overactive, they drive excessive proliferation. Tumour suppressor genes normally restrain cell division or promote apoptosis, and their inactivation removes these brakes.
A well-known tumour suppressor is p53, which is mutated in more than half of all human cancers. A well-known oncogene is RAS, which encodes a signalling protein; mutations in RAS keep the protein permanently active, continuously stimulating cell division. Typically, cancer develops only after several mutations accumulate, which explains why cancer incidence increases with age.
