📚 DNA Structure and Alleles | DNA
Deoxyribonucleic acid, or DNA, is the remarkable molecule that carries the genetic blueprint for every living organism. It stores the instructions needed to build proteins, control cellular activities, and pass traits from one generation to the next. Variations in these instructions arise from different forms of genes called alleles, which are the foundation of inheritance and explain why offspring resemble but are not identical to their parents. Understanding the double-helix structure of DNA and the concept of alleles is therefore central to modern biology and a core topic in the Edexcel IGCSE Science specification.
1. What is DNA? | 什么是DNA?
DNA stands for deoxyribonucleic acid. It is a long polymer made up of repeating units called nucleotides. In eukaryotic cells, such as those of animals and plants, most DNA is located inside the nucleus, organised into structures known as chromosomes. DNA contains the coded information that determines an organism's characteristics, from eye colour to enzyme production.
2. The Discovery of the Double Helix |
The three-dimensional structure of DNA was determined in 1953 by James Watson and Francis Crick, who built on the X-ray crystallography data produced by Rosalind Franklin and the work of Maurice Wilkins. Watson and Crick proposed that DNA consists of two polynucleotide strands coiled around each other to form a double helix. This breakthrough revolutionised genetics, earning Watson, Crick, and Wilkins the Nobel Prize in 1962.
3. The Building Blocks: Nucleotides |
Each nucleotide in DNA is composed of three parts: a phosphate group, a deoxyribose sugar, and a nitrogenous base. The phosphate and sugar link together to form the backbone of each DNA strand, while the base sticks out to the side. There are four different bases in DNA: adenine (A), thymine (T), cytosine (C), and guanine (G).
- Phosphate group: Provides the link between sugars of adjacent nucleotides, forming a strong sugar–phosphate backbone. |
- Deoxyribose sugar: A pentose sugar that gives DNA its name (deoxyribonucleic acid).
- Nitrogenous base: One of four molecules—A, T, C, G—that carry the genetic code.
4. Complementary Base Pairing |
The two strands of DNA are held together by hydrogen bonds between complementary base pairs. Adenine always pairs with thymine via two hydrogen bonds, and cytosine always pairs with guanine via three hydrogen bonds. This complementary rule ensures that the sequence on one strand dictates the sequence on the other, enabling accurate DNA replication.
A = T (2 hydrogen bonds) | C ≡ G (3 hydrogen bonds)
A = T(2个氢键) | C ≡ G(3个氢键)
5. The Double Helix Shape |
DNA's two antiparallel strands twist into a right-handed double helix. The sugar–phosphate backbones run along the outside, protecting the bases, while the base pairs lie stacked on the inside, like the steps of a spiral staircase. This compact and stable structure safeguards the genetic code and allows the molecule to be densely packed into chromosomes.
6. From DNA to Genes | 从DNA到基因
A gene is a specific section of DNA that contains the instructions for making a particular protein. The sequence of bases along the gene determines the order of amino acids in the protein. Humans possess around 20,000–25,000 genes distributed across 46 chromosomes. Each chromosome is a single, very long DNA molecule wrapped around histone proteins.
7. Introducing Alleles |
Genes often exist in slightly different forms called alleles. An allele is one variant of a gene that arises by mutation and occupies the same position, or locus, on a chromosome. For example, the gene controlling human earlobe attachment has two common alleles: one for free earlobes and one for attached earlobes. Alleles are responsible for the subtle differences between individuals of the same species.
8. Dominant and Recessive Alleles |
Alleles can be dominant or recessive. A dominant allele always expresses its characteristic even if only one copy is present, whereas a recessive allele is expressed only when two copies are present (homozygous recessive). In genetic diagrams, dominant alleles are conventionally represented by uppercase letters and recessive alleles by lowercase letters, such as 'E' for free earlobes (dominant) and 'e' for attached earlobes (recessive).
|---|---|
| EE | Free earlobes |
| Ee | Free earlobes |
9. Genotype, Phenotype, and Allele Combinations |
The genotype refers to the combination of alleles an organism has for a particular gene, while the phenotype is the visible outcome or characteristic. If the two alleles are identical, the individual is homozygous for that trait; if they are different, the individual is heterozygous. In a heterozygous individual, only the dominant allele influences the phenotype.
10. Predicting Inheritance Using Punnett Squares |
nett square predicts a genotype ratio of 1 EE : 2 Ee : 1 ee and a phenotype ratio of 3 free earlobes : 1 attached earlobe. This pattern explains why recessive traits can skip a generation.
Punnett Square for Ee × Ee: offspring genotypes = 25% EE, 50% Ee, 25% ee
11. Alleles and Genetic Variation |
Multiple alleles can exist for a single gene within a population, though each individual carries only two alleles (one from each parent). A classic example is the ABO blood group system, where three alleles—IA, IB, and i—give rise to four blood types. Such allelic diversity is the raw material for natural selection and evolution, contributing to the rich variety of life on Earth.
12. Connecting Structure to Function |
The elegance of DNA's double-helix structure is intimately tied to its roles in protein synthesis and inheritance. The sequence of bases along a gene dictates the order of amino acids; any change in that sequence—a mutation—can create a new allele. Over generations, mutations and sexual reproduction shuffle alleles, producing the continuous variation that allows populations to adapt. Thus, understanding DNA and alleles is foundational not only for genetics but for the entire study of life sciences.
