📚 Substitution Reactions |
In organic chemistry, substitution reactions are among the most fundamental transformations, allowing one atom or group in a molecule to be replaced by another. These reactions are not only central to synthetic pathways but also reveal deep mechanistic insights into how molecules interact. From the chlorination of methane to the nitration of benzene and the hydrolysis of halogenoalkanes, substitution reactions appear across the IB Chemistry syllabus, linking topics such as bonding, kinetics, and organic synthesis.
1. Introduction to Substitution Reactions |
nd is replaced by another atom, ion, or group. The general equation can be summarised as R–X + Y → R– chanism followed. In IB Chemistry, we primarily study three major types: free radical substitution, electrophilic aromatic substitution, and nucleophilic substitution.
Understanding substitution mechanisms helps chemists predict products, control reaction conditions, and design synthetic routes. The IB syllabus expects students to describe mechanisms using curly arrows, identify rate–determining steps, and relate molecular structure to reactivity. While HL students delve deeper into SN1 and SN2 kinetics and stereochemistry, SL students focus on recognising patterns and drawing simple mechanisms.
2. Types of Substitution Reactions |
Substitution reactions are broadly divided according to the reaction mechanism. The three main categories covered in IB are free radical substitution, electrophilic substitution (especially in benzene derivatives), and nucleophilic substitution (SN1 and SN2). Each type is characterised by a distinct pattern of bond breaking and bond making. Free radical substitution involves the homolytic cleavage of covalent bonds, generating reactive radical intermediates. Electrophilic substitution proceeds via an electrophile attacking an electron–rich system, typically an aromatic ring. Nucleophilic substitution features a nucleophile donating a pair of electrons to an electron–deficient carbon centre.
Additionally, it is crucial to distinguish between the terms reagent, substrate, and leaving group. The substrate is the molecule that undergoes substitution (e.g., a halogenoalkane), the reagent supplies the incoming atom or group, and the leaving group is the species displaced during the reaction. The interplay between nucleophilicity, electrophilicity, and leaving group ability determines the pathway and rate of a given reaction.
3. Free Radical Substitution: Mechanism |
Free radical substitution occurs when a molecule with a relatively weak covalent bond, such as a halogen, undergoes homolytic fission in the presence of ultraviolet (UV) light or heat. The reaction proceeds via a chain mechanism comprising three stages: initiation, propagation, and termination. Initiation generates free radicals by breaking a bond symmetrically; each atom takes one electron from the bond pair, producing two uncharged species with an unpaired electron, denoted as radicals.
In the propagation steps, a radical reacts with a stable molecule to form a new radical and a stable product, thus continuing the chain. The initial radical abstracts a hydrogen atom from an alkane, creating an alkyl radical. This alkyl radical then reacts with a halogen molecule to generate the halogenated product and a new halogen radical. Termination occurs when any two radicals combine, consuming the radical species and ending the chain reaction.
4. Free Radical Substitution: Example of Methane Chlorination |
r substitution products forming if chlorine is in excess. The initiation step is Cl₂ → 2Cl• d. In the first propagation step, a chlorine radical abstracts a hydrogen from methane: Cl• + CH₄ → HCl + •CH₃. The methyl radical then attacks a chlorine molecule: •CH₃ + Cl₂ → CH₃Cl + Cl• second propagation step regenerates the chlorine radical, allowing the chain to continue.
Termination possibilities include the combination of two chlorine radicals (Cl• + Cl• → Cl₂), two methyl radicals (•CH₃ + •CH₃ → C₂H₆), or a chlorine and a methyl radical (Cl• + • mation of trace amounts of ethane, which is evidence for the existence of methyl radicals.
5. Electrophilic Substitution in Benzene |
Benzene, with its delocalised pi electron system, is highly attractive to electrophiles. However, because the aromatic ring is stabilised by resonance, benzene does not undergo typical addition reactions; instead, it undergoes electrophilic substitution, where one hydrogen atom is replaced by an electrophile while the aromaticity is preserved. The general mechanism involves an electrophile, E⁺, generated in situ by a catalyst, attacking the pi system to form a carbocation intermediate (sometimes called a Wheland complex or arenium ion) and then losing a proton to regenerate the stable aromatic ring.
In the IB syllabus, students are required to know two main electrophilic substitution reactions of benzene: nitration and halogenation. Both require a strong electrophile and a catalyst. The mechanism is identical in principle for other electrophilic substitutions such as Friedel–Crafts alkylation and acylation, though these are often discussed at HL. The delocalised model of benzene is essential for explaining why substitution occurs rather than addition.
6. Nitration of Benzene: Mechanism |
The nitration of benzene introduces a nitro group (– f the electrophile: HNO₃ + H₂SO₄ → NO₂⁺ + HSO₄⁻ + H₂O. The nitronium ion is a powerful electrophile that attacks the benzene ring.
HNO₃ + H₂SO₄ → NO₂⁺ + HSO₄⁻ + H₂O。
In the substitution step, the pi electrons of benzene are donated to the electron–deficient nitrogen of NO₂⁺, forming a positively charged intermediate. The ring temporarily loses aromaticity and bears a positive charge, delocalised over several carbon atoms. This intermediate then loses a proton (H⁺) from the carbon that was attacked, restoring the aromatic sextet and yielding nitrobenzene. The proton is taken up by HSO₄⁻ to regenerate H₂SO₄, completing the catalytic cycle.
7. Halogenation of Benzene |
Benzene can be chlorinated or brominated using a halogen in the presence of a Lewis acid catalyst such as AlCl₃ or FeBr₃. The catalyst polarises the halogen molecule, generating a more powerful electrophile. For bromination, FeBr₃ is often generated in situ from iron filings and bromine. The electrophile is the Br⁺–FeBr₄⁻ complex, effectively acting as Br⁺. The mechanism follows the same electrophilic substitution pathway: the electrophile is attacked by the benzene pi system, a carbocation intermediate forms, and then rapid deprotonation restores aromaticity, giving bromobenzene and HBr.
Halogenation of benzene is more challenging than alkene addition because the aromatic ring is stabilised. A strong electrophile is essential, and the presence of the catalyst is critical. Without a catalyst, the reaction between benzene and bromine is extremely slow. In the IB HL course, students should be able to draw the mechanism with curly arrows showing the movement of electron pairs from the ring to the electrophile and then the loss of a proton.
8. Nucleophilic Substitution: General Features |
Nucleophilic substitution (SN) reactions involve the replacement of a leaving group on a saturated carbon atom by a nucleophile. The nucleophile is an electron pair donor seeking a positive or partially positive centre. In IB Chemistry, SN reactions are studied mainly with primary, secondary, and tertiary halogenoalkanes, where the leaving group is a halide ion such as Cl⁻, Br⁻, or I⁻. Hydroxide ions (OH⁻), cyanide ions (CN⁻), and ammonia (NH₃) are common nucleophiles.
The general equation is Nu:⁻ + R–X → R–Nu + X:⁻. Two distinct mechanisms exist: SN1 and SN2. The 'SN' nds for substitution nucleophilic, while the number refers to the molecularity of the rate– te). The choice of mechanism depends on the structure of the substrate, the nature of the nucleophile, the leaving group, and the solvent.
9. SN1 Mechanism | SN1机理
The SN1 mechanism proceeds in two steps. The first, rate–determining step is the heterolytic cleavage of the carbon–halogen bond, forming a planar carbocation intermediate and the halide ion. This step is slow and depends only on the concentration of the halogenoalkane. The second step is the rapid attack of the nucleophile on the electron–deficient carbocation, resulting in product formation. Because the carbocation is planar, the nucleophile can attack from either face, leading to a racemic mixture if the starting carbon is chiral.
SN1 is favoured by tertiary halogenoalkanes because the resulting carbocation is stabilised by the electron–donating inductive effect of three alkyl groups. Secondary substrates can also react via SN1, especially in polar protic solvents that stabilise the carbocation and the leaving group through solvation. Primary substrates rarely undergo SN1 due to the instability of primary carbocations. SN1 reactions often show mixed stereochemistry and can be accompanied by elimination (E1) as a side reaction under basic conditions.
10. SN2 Mechanism | SN2机理
The SN2 mechanism is a one– sly as the new bond forms. The reaction is bimolecular, with the rate law Rate = k[Nu:⁻][R– ration at the carbon centre, analogous to an umbrella turning inside out in a strong wind.
SN2 is favoured by primary substrates due to minimal steric hindrance around the reaction centre. Secondary substrates also undergo SN2, but at a slower rate. Tertiary halogenoalkanes are essentially inert toward SN2 because the backside attack is sterically blocked by the three bulky alkyl groups. A strong nucleophile and an aprotic polar solvent further promote the SN2 pathway by keeping the nucleophile highly reactive and unencumbered. Leaving group ability also affects SN2 rates: good leaving groups such as I⁻ and tosylate accelerate the reaction.
Several factors determine whether a nucleophilic substitution will follow the SN1 or SN2 pathway. Substrate structure is the most critical: tertiary substrates strongly favour SN1, primary substrates favour SN2, and secondary can proceed via either depending on conditions. Nucleophile strength primarily affects SN2; strong nucleophiles push the reaction toward the concerted path. For SN1, nucleophile strength is far less significant because the nucleophile is not involved in the slow step.
Leaving group ability influences both mechanisms. Good leaving groups are weak bases that can stabilise the negative charge after departure. Iodide is an excellent leaving group, whereas fluoride is poor. The solvent also plays a decisive role: polar protic solvents like water or alcohols stabilise the carbocation and the leaving group, thus accelerating SN1. Polar aprotic solvents, such as propanone, solvate cations strongly but leave anions relatively free, enhancing nucleophilicity for SN2. Temperature and concentration can also shift the preference, but the substrate structure remains the main determining factor.
12. Summary and Comparison |
Substitution reactions unify large areas of organic chemistry by showing how functional groups can be interconverted. Free radical substitution enables functionalisation of alkanes, though with limited selectivity. Electrophilic aromatic substitution is the key method for introducing substituents onto benzene rings without disrupting aromaticity. Nucleophilic substitution, through the SN1 and SN2 mechanisms, is essential for building complex molecules from halogenoalkanes and related substrates.
A comparison of SN1 and SN2 highlights key differences: SN1 is stepwise, proceeds with racemisation, favours tertiary carbons, and its rate is independent of nucleophile concentration; SN2 is concerted, proceeds with inversion, favours primary carbons, and is second–order overall. In electrophilic substitution, the aromatic ring retains its stability throughout, whereas in free radical substitution, highly reactive intermediates drive the chain. Mastering these mechanisms allows IB students to rationalise reaction outcomes, predict products, and understand kinetic and stereochemical data, solidifying their command of organic chemical principles.
