1. Cell Biology and Microscopy |

All organisms are composed of cells, the fundamental units of life. Cells can be prokaryotic (e.g., bacteria) lacking a true nucleus, or eukaryotic (e.g., plants, animals, fungi) possessing a membrane-bound nucleus that encloses genetic material. Prokaryotic cells are much smaller, usually 0.2-2 μm across, and carry a single circular loop of DNA plus small rings called plasmids.

Animal and plant cells share common organelles: a nucleus that controls activities, mitochondria where aerobic respiration occurs, ribosomes for protein synthesis, and cytoplasm as the site of chemical reactions. Plant cells additionally have a rigid cellulose cell wall, a large permanent vacuole, and chloroplasts containing chlorophyll for photosynthesis.

solution but kill the specimen. When calculating magnification, convert all lengths to the same unit first: 1 mm = 1000 μm and 1 μm = 1000 nm.

Magnification = Image size ÷ Actual size

2. Enzymes and Transport Across Membranes |

Enzymes are biological catalysts made of protein. They speed up reactions by lowering the activation energy needed, and they are specific because the substrate must fit the active site, the so-called lock and key model. The enzyme is not used up and can be reused many times.

Temperature and pH affect enzyme activity. Each enzyme has an optimum; human enzymes usually work best near 37 °C and pH 7, while stomach protease works best around pH 2. Beyond the optimum, bonds break, the active site changes shape and the enzyme becomes denatured, so activity falls sharply and never recovers.

Substances cross membranes by diffusion, osmosis or active transport. Diffusion is the net movement of particles from high to low concentration, driven by the concentration gradient and affected by temperature, surface area and diffusion distance. Osmosis is the diffusion of water through a partially permeable membrane from a dilute solution to a concentrated one.

Active transport moves substances against the concentration gradient and therefore requires energy from respiration plus carrier proteins in the membrane. Root hair cells use it to absorb mineral ions, and the small intestine uses it to absorb glucose when gut concentration is lower than blood concentration.

Rate of reaction = 1 ÷ Time taken

3. Cell Division, Stem Cells and Chromosomes |

Body cells divide by mitosis to produce two genetically identical diploid daughter cells, each with the same chromosome number as the parent. In humans this is 46 chromosomes, or 23 pairs. Before division, the DNA is replicated in interphase so that each chromosome consists of two identical chromatids.

Mitosis is needed for growth, repair of damaged tissue and asexual reproduction. Bacteria divide by a simpler process called binary fission, which can occur every 20 minutes in ideal conditions and explains how infections spread rapidly inside the body.

Stem cells are unspecialised cells that can divide and differentiate into specialised cells such as nerve, muscle or red blood cells. Embryonic stem cells can become almost any cell type, adult stem cells are more limited, and plant meristems at shoot and root tips provide a lifelong supply of new cells.

Possible medical uses include replacing damaged tissue in diabetes, Parkinson disease and spinal injury, but ethical issues arise from using embryos. Cancer is the result of uncontrolled mitosis caused by mutations in genes controlling the cell cycle, and tumours form when these cells keep dividing.

4. Photosynthesis and Respiration |

Photosynthesis is an endothermic reaction in which green plants, algae and some bacteria convert light energy into chemical energy stored in glucose. It occurs mainly in the palisade cells of leaves, which contain many chloroplasts and are packed near the upper surface.

6CO₂ + 6H₂O → C₆H₁₂O₆ + 6O₂

The rate of photosynthesis is limited by light intensity, carbon dioxide concentration and temperature. Whichever factor is in shortest supply becomes the limiting factor, and increasing the others will not raise the rate until that factor is also increased. Temperature is limiting because the reactions are controlled by enzymes.

Practical work often uses pondweed, counting oxygen bubbles released per minute at different distances from a lamp, or measuring the volume of gas collected. Greenhouses use this knowledge by adding carbon dioxide, artificial light and heaters to increase crop yield.

Aerobic respiration releases much more energy and takes place mainly in the mitochondria.

C₆H₁₂O₆ + 6O₂ → 6CO₂ + 6H₂O + Energy

Anaerobic respiration happens without oxygen. In muscles, glucose is converted to lactic acid, which builds up and causes fatigue and cramp; the extra oxygen needed to break it down afterwards is called the oxygen debt. In yeast and plant cells, glucose is converted to ethanol and carbon dioxide, a process used in bread making and brewing.

5. Biological Molecules and Food Tests |

Living organisms are built from four main groups of biological molecules: carbohydrates, proteins, lipids and nucleic acids. Carbohydrates such as starch and glycogen are made of simple sugars and act as energy stores; proteins are polymers of amino acids used for growth and repair; lipids store energy and form cell membranes.

Food tests identify these molecules quickly. Starch turns iodine solution blue-black, reducing sugars turn Benedict solution from blue through green and yellow to brick red after heating in a water bath, protein turns Biuret reagent purple, and lipids form a white emulsion when shaken with ethanol and water.

TestReagentPositive Result
StarchIodine solutionBlue-black
Reducing sugarBenedict solutionBrick red precipitate
ProteinBiuret reagentPurple
LipidEthanol + waterWhite emulsion

Energy requirements vary with age, sex and activity level. A diet too high in energy leads to obesity, which raises the risk of type 2 diabetes and cardiovascular disease, while a diet too low in energy causes malnutrition. Body mass index and waist-to-hip ratio are simple measures used to assess body composition.

6. Digestion and the Digestive System |

Digestion converts large insoluble molecules into small soluble ones that can be absorbed into the blood. Mechanical digestion begins in the mouth, where teeth break food into smaller pieces to increase the surface area for enzymes, and the tongue forms a bolus for swallowing.

Three main groups of digestive enzymes work in the gut. Carbohydrase, including amylase from salivary glands and the pancreas, breaks starch into maltose and then glucose. Protease from the stomach, pancreas and small intestine breaks protein into amino acids. Lipase from the pancreas breaks lipids into fatty acids and glycerol.

Bile is produced by the liver, stored in the gall bladder, and released into the small intestine. It is alkaline, so it neutralises hydrochloric acid from the stomach and provides the optimum pH for pancreatic enzymes. It also emulsifies lipids into tiny droplets, increasing surface area for lipase.

The small intestine is adapted for absorption with villi and microvilli that greatly increase surface area, a wall only one cell thick for a short diffusion path, and a dense network of capillaries that carries absorbed nutrients away and maintains a steep concentration gradient.

7. Transport Systems: Blood, Heart and Plants |

Blood is a tissue made of plasma, red blood cells, white blood cells and platelets. Red blood cells have no nucleus, are biconcave in shape and contain haemoglobin, which binds oxygen in the lungs and releases it in respiring tissues. White blood cells defend the body, while platelets help blood clot at wounds.

The heart has four chambers and a double circulatory system. Deoxygenated blood returns to the right atrium, passes to the right ventricle and is pumped to the lungs; oxygenated blood returns to the left atrium and is pumped by the thicker-walled left ventricle to the whole body. Valves prevent backflow.

Coronary heart disease is caused by fatty deposits narrowing the coronary arteries, reducing oxygen supply to heart muscle. Treatments include stents to hold arteries open, statins to lower cholesterol, and lifestyle changes such as stopping smoking and taking regular exercise. Faulty valves can be replaced, and an artificial pacemaker can correct irregular heart rhythm.
